Design of freeze-drying cycles: The determination of heat transfer coefficient by using heat flux sensor and MicroFD

Marco Carfagna et al.

Summary: This study investigates the combination use of heat flux sensor (HFS) and miniaturized freeze-dryer (MicroFD) to obtain the heat transfer coefficient (Kv) for different formulation types and freezing protocols. The results show that this combination approach can provide real-time Kv with less effort compared to traditional gravimetric method, and can be used to study different vial scenarios and design lyophilization processes.

Advanced Process Analytical Technology in Combination with Process Modeling for Endpoint and Model Parameter Determination in Lyophilization Process Design and Optimization

Alex Juckers et al.

Summary: Lyophilization is commonly used for preserving thermolabile products, with the main drawback being the long processing time. By implementing the quality by design (QbD) approach and process analytical technology (PAT), the process duration can be optimized for increased productivity while maintaining product safety.

Applicability of Raman and near-infrared spectroscopy in the monitoring of freeze-drying injectable ibuprofen

Maja Preskar et al.

Summary: The study demonstrates the feasibility of using NIR and Raman spectroscopy for monitoring the freeze-drying process, with NIR being a useful tool for predicting residual moisture content and IBP values. Despite some challenges with overlapping absorption bands in a multi-component system, NIR spectra were consistent with HPLC measurements.
