Safety study of Rift Valley Fever human vaccine candidate (DDVax) in mosquitoes

Summary: The study showed that DDVax had significantly lower infection rates in mosquitoes and could not be transmitted between mosquitoes. Although infectious virus could be isolated from the bodies of mosquitoes fed on goats inoculated with DDVax or MP-12, the number of DDvax-positive mosquitoes was significantly reduced. Furthermore, DDVax did not disseminate to the legs and wings of mosquitoes fed on goats.

Summary: RVF, a priority zoonotic disease, can be replicated in Vero cells to generate recombinant virus. However, reamplification of virus clones in Vero cells might increase the subpopulations of mutants. The strategy of rescuing recombinant RVFV in Vero cells and reamplifying in MRC-5 cells will support the production of vaccines for human use.
