Crystal Engineering of Pharmaceutical Cocrystals in the Discovery and Development of Improved Drugs

Summary: Crystal engineering is a discipline that started in the 1970s, focusing on the study of topochemical reactions in the solid state. One important application of crystal engineering is in the design of pharmaceutical cocrystals, which can systematically tune the physicochemical properties of drugs without altering their molecular structure. This review traces the development of supramolecular synthons as robust hydrogen bond patterns for pharmaceutical cocrystal design, and discusses recent advancements in the field that have led to the launch of several drugs with improved efficacy and bioavailability. Polymorphism in Ionic Cocrystals Comprising Lithium Salts and L-Proline

Novel cocrystals of itraconazole: Insights from phase diagrams, formation thermodynamics and solubility

Summary: In this study, the cocrystallization of itraconazole with 4-aminobenzoic acid and 4-hydroxybenzamide led to the formation of two new solid forms, with detailed analysis on their stoichiometry, polymorphic forms, and dissolution behavior. The cocrystallization with 4OHBZA was found to significantly increase the drug solubility and exhibited higher AUC values compared to the drug.

Celecoxib-tramadol co-crystal: A Randomized 4-Way Crossover Comparative Bioavailability Study

The in situ generated N-heterocyclic carbene from azolium salt is trapped by Palladium ion leading to NHC-Palladium complexes. NHCs behaves as an efficient nucleophilic catalyst with strong sigma-donating character, due to this it can easily bind with a range of metallic and non-metallic centre.

This review provides updates on 61 pharmaceutical salts approved by the FDA between 2015 and 2019, detailing their chemical structures and therapeutic indications.
