Claudin-10a Deficiency Shifts Proximal Tubular Cl- Permeability to Cation Selectivity via Claudin-2 Redistribution

Summary: Recent study indicates that claudin-10a is the major paracellular anion channel in the proximal tubule, and its deficiency leads to excessive reabsorption of calcium and magnesium. Various analyses, including electrophysiological studies, suggest compensatory transcellular transport in proximal and distal tubule segments, as well as metabolic adaptation in the proximal tubule, to counterbalance the loss of paracellular anion permeability.

Computational Assessment of Different Structural Models for Claudin-5 Complexes in Blood-Brain Barrier Tight Junctions

Computational study of ion permeation through claudin-4 paracellular channels

Summary: Claudins are a family of protein homologs that play a crucial role in cell-to-cell tight junctions. Despite experimental evidence suggesting that Claudin 4 can form anion-selective channels in the renal epithelium, its exact function remains unclear. This study uses computational methods to transfer structural models of Claudin-based complexes to Claudin 4 and investigates the thermodynamic properties driving ion selectivity. The results show that one model allows chloride permeability and repels cations, while the other forms a barrier to the passage of major physiological ions. Additionally, the study confirms the charge selectivity role of the residue Lys65 in regulating paracellular permeability.

Role of TM3 in claudin-15 strand flexibility: A molecular dynamics study

Summary: In this study, the behavior of claudin-15 strands within lipid membranes and the effect of a single-point mutation on the morphology of the strands were investigated using molecular dynamics. The results showed that the mutation significantly altered the lateral flexibility of the strands, resulting in a three-fold increase in persistence length. The mutation introduced a bend in the third transmembrane helix, which restricted the fluctuations of the strands and led to higher persistence lengths.
