Elucidation of structure-function relationships in Methanocaldococcus jannaschii RNase P, a multi-subunit catalytic ribonucleoprotein

Hong-Duc Phan et al.

Summary: RNase P is a ribonucleoprotein that removes the 5' leader from precursor tRNAs. The study reveals the dimeric configuration of Mja RNase P, with each monomer containing one RPR and five RPPs. It confirms the oligomeric state of Mja RNase P and shows the different stoichiometry of each monomer, with up to two copies of L7Ae. Disrupting the L7Ae-RPR interaction does not affect the assembly and function of RNase P due to redundancy in protein-protein interactions.

RPP30 is a novel diagnostic and prognostic biomarker for gastric cancer

Ying Kan et al.

Summary: This study aimed to identify the hub gene in gastric cancer tumorigenesis and constructed a biomarker prediction model. It was found that high RPP30 expression in gastric cancer was significantly correlated with disease progression and poor survival, promoting tumorigenesis and angiogenesis via tRNA dysregulation.

Cancer in Systemic Sclerosis: Analysis of Antibodies Against Components of the Th/To Complex

Christopher A. Mecoli et al.

Summary: This study aimed to describe the prevalence and clinical characteristics of autoantibodies against components of the Th/To complex in SSc patients and determine if they are associated with cancer. Patients with these antibodies showed a clinical phenotype of limited cutaneous disease and pulmonary involvement, and were less likely to develop cancer within 2 years of SSc onset. The findings suggest that Th/To autoantibodies may have a protective effect against contemporaneous cancer.

The rice RNase P protein subunit Rpp30 confers broad-spectrum resistance to fungal and bacterial pathogens

Wei Li et al.

Summary: OsRpp30, functioning as a catalytic ribonucleoprotein RNP subunit in rice, plays multiple roles including chromatin regulation and innate immunity. It is shown to increase expression post-infection, co-localize with HDT701 in the nucleus, and enhance defense gene expression and ROS production, resulting in resistance to fungal and bacterial pathogens in rice. OsRpp30, conserved in grass crops, co-purifies with RNase P pre-tRNA cleavage activity and has a crucial role in defense regulation, suggesting its potential for generating disease-resistant rice cultivars.

Novel NPM1 exon 5 mutations and gene fusions leading to aberrant cytoplasmic nucleophosmin in AML

Maria Paola Martelli et al.

Summary: This study identified mutations in Nucleophosmin (NPM1) in acute myeloid leukemia (AML) affecting not only exon 12, but also exons 9, 11, and even exon 5. Novel rearrangements of NPM1 were also discovered. Functional studies showed the importance of cytoplasmic accumulation of NPM1 in leukemogenesis. Additional assays for molecular diagnosis of NPM1-mutated AML are needed.

Aileen G. Rowan et al.

Summary: Quantifying the SARS-CoV-2 viral copy number is crucial for clinical and public health management. A duplex RT-qPCR assay was shown to be effective in clinical settings and the internal control revealed inadequacies in some samples. This quantification method provides valuable information for decision making.
