Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Inherited TP53 Variants and Risk of Prostate Cancer

Kara N. Maxwell et al.

Summary: Complementary analysis of prostate cancer incidence in LFS males and gTP53 prevalence in prostate cancer cohorts suggests that gTP53 predisposes to aggressive prostate cancer. Prostate cancer should be considered as part of LFS screening protocols and TP53 should be considered in germline prostate cancer susceptibility testing.

Enhanced autophagy and NFE2L2/NRF2 pathway activation in SPOP mutation-driven prostate cancer

Kun Gao et al.

Summary: This study reveals a negative regulatory role of SPOP in autophagy and NFE2L2 pathway activation. The study found that SPOP induces non-degradative ubiquitination of SQSTM1, thereby suppressing autophagy and NFE2L2 pathway activation. The PCa-associated SPOP mutants lose the capacity to ubiquitinate SQSTM1 and instead enhance autophagy and the antioxidant response in a dominant-negative manner.

UHRF1 promotes androgen receptor-regulated CDC6 transcription and anti-androgen receptor drug resistance in prostate cancer through KDM4C-Mediated chromatin modifications

Yingxue Gao et al.

Summary: UHRF1 and CDC6, oncogenes that play critical roles in therapeutic resistance, are upregulated simultaneously in prostate cancer, where UHRF1 promotes CDC6 transcription controlled by AR through chromatin modification, contributing to anti-AR drug resistance and worse survival. Targeting both AR and UHRF1 simultaneously could be a novel and promising therapeutic approach for prostate cancer.

DNMT3A epigenetically regulates key microRNAs involved in epithelial-to-mesenchymal transition in prostate cancer

Monica Mancini et al.

Summary: The study demonstrates that the interaction between DNMT3A and miRNAs plays a central role in the EMT process of prostate cancer, with key miRNAs such as the miR-200 family, miR-205, and miR-203 being down-regulated and hyper-methylated. Differential expression of DNMT3A isoforms and a regulatory feedback loop between miR-429 and DNMT3A were also identified during EMT. Additionally, miR-429 was shown to target DNMT3A and modulate the expression of EMT factors.

The Mutational Landscape of Metastatic Castration-sensitive Prostate Cancer: The Spectrum Theory Revisited

Matthew P. Deek et al.

Summary: Somatic mutational profiles across the spectrum of metastatic castration-sensitive prostate cancer reveal associations between TP53 mutations and clinical outcomes in patients with oligometastasis, indicating a potential biological definition of oligometastasis beyond numerical classifications. Mutation analysis in TP53 and DNA double-strand break repair genes may help predict the number of metastases and clinical outcomes in patients with mCSPC.

Epigenetic modulation of antitumor immunity for improved cancer immunotherapy

Enyong Dai et al.

Summary: Epigenetic mechanisms play important roles in cancer initiation and immune cell function, with the potential to impact tumor microenvironment and therapeutic efficacy by modulating immune cell populations and promoting transcriptional and metabolic reprogramming. By regulating immune-associated genes and immune checkpoint molecules, epigenetic modulating agents can enhance tumor immunogenicity and facilitate anti-tumor immune responses.

Analysis of RNA m6A methylation regulators and tumour immune cell infiltration characterization in prostate cancer

Yue Zhao et al.

Summary: Recent studies have shown the potential roles of RNA N6-methyladenosine (m(6)A) modification in tumor microenvironment (TME) cell infiltration. The expression of different m(6)A regulators within prostate cancer is related to genetic variation, alternative splicing, tumor mutational burden (TMB) and TME, and can predict TME scores and prognosis. CD8 T cells play a crucial role in immune cell intersection and are strongly correlated with immune associated gene sets in prostate cancer.
