AAV-mediated delivery of an anti-BACE1 VHH alleviates pathology in an Alzheimer's disease model

Summary: Single domain antibodies (VHHs) have potential therapeutic value for neurological disorders, but their use in the central nervous system is limited by the blood-brain barrier. This study proposes a gene transfer strategy using BBB-crossing AAV-based vectors to deliver VHH directly into the CNS. The results show that AAV-delivered VHH can effectively inhibit BACE1 and improve cognitive performance in an Alzheimer's disease mouse model, providing a novel therapeutic approach for neurodegenerative diseases.

TDP-43 represses cryptic exon inclusion in the FTD-ALS gene UNC13A

Summary: A hallmark feature of ALS and FTD is the depletion of RNA-binding protein TDP-43 from neurons, and a major function of TDP-43 is to repress cryptic exon inclusion. UNC13A gene polymorphisms are strongly associated with FTD and ALS, and this study shows that loss of TDP-43 leads to cryptic exon splicing in UNC13A. The top genetic risk factors for FTD and ALS (UNC13A genetic variants) are shown to be functionally linked to TDP-43 dysfunction.

Chimeric cerebral organoids reveal the essentials of neuronal and astrocytic APOE4 for Alzheimer's tau pathology

Summary: Researchers developed a novel engineering method to assess the role of apolipoprotein E4 (APOE4) in neurons and astrocytes using chimeric human cerebral organoids (chCOs). The results showed that APOE4 promoted lipid droplet formation and cholesterol accumulation in neurons, and also affected levels of tau protein and A beta. This study revealed the essence of APOE4 in both neurons and astrocytes for tau pathology, and suggested that chCOs could serve as a valuable pathological model for Alzheimer's disease (AD) research and drug discovery.

Human ALS/FTD brain organoid slice cultures display distinct early astrocyte and targetable neuronal pathology

Neurodevelopmental defects and neurodegenerative phenotypes in human brain organoids carrying Parkinson's disease-linked DNAJC6 mutations

The defects in DNAJC6-mediated endocytosis impair the WNT-LMX1A signal during mDA neuron development, leading to the generation of vulnerable neurons with pathologic manifestations.
