Tumor-Infiltrating Dendritic Cells: Decisive Roles in Cancer Immunosurveillance, Immunoediting, and Tumor T Cell Tolerance

Hypoxia-driven immunosuppression by Treg and type-2 conventional dendritic cells in HCC

Summary: This study analyzed the immune landscapes of hypoxic regions in hepatocellular carcinoma (HCC) and identified enriched immune subsets, including regulatory T cells, M2 macrophages, and HLA-DRlo cDC2, in hypoxia-high tumor regions. In contrast, hypoxia-low tumor regions showed an abundance of active granzyme B-hi PD-1(lo) CD8(+) T cells, indicating a relatively active immune landscape. The up-regulation of cancer-associated genes in tumor tissues and immunosuppressive genes in tumor-infiltrating leukocytes supported a highly pro-tumorigenic network in hypoxic HCC.

Amateur antigen-presenting cells in the tumor microenvironment

Recent evidence suggests that besides professional antigen-presenting cells, other cells in the tumor microenvironment are also capable of presenting tumor antigens, potentially playing a significant role in priming an antitumor immune response. This new perspective on antigen presentation supports the development of novel immunotherapies to enhance T cell priming.

Quiescent cancer cells resist T cell attack by forming an immunosuppressive niche

Summary: This study discovered that in primary breast cancer, tumor cells that resist T cell attack are quiescent and form clusters with reduced immune infiltration. These quiescent cancer cells show superior tumorigenic capacity and higher expression of chemotherapy resistance and stemness genes. Single-cell RNA sequencing revealed differential phenotypes in infiltrating cells based on their intra-tumor location, and uncovered hypoxia-induced immune-suppressive programs. The findings suggest that eliminating quiescent cancer cells holds the promise to counteract immunotherapy resistance and prevent disease recurrence in TNBC.

The composition of TIMs appeared to be linked with certain features of somatic mutations and gene expressions.

Recent studies have found that cross-priming of conventional type 1 dendritic cells (cDC1) plays a crucial role in initiating CD8(+) T-cell responses during tumor progression, as well as in immunotherapy-mediated reactivation of tumor-specific CD8(+) T cells for tumor regression. Inducing type I IFNs within tumors can overcome innate immune resistance and activate antitumor adaptive immunity.
