Astrocyte regulation of synaptic signaling in psychiatric disorders

Summary: Neuroscience has recognized the important role of astroglia in neuronal synaptic activity and considers them as an equal partner in tripartite synaptic transmission and plasticity. However, the lack of reagents and technologies for studying astroglia has hindered the understanding of tripartite synaptic physiology. This article reviews the role of astroglia in synaptic function and behavior regulation, and explores their impact on psychiatric disorders.

Astrocyte Molecular Clock Function in the Nucleus Accumbens Is Important for Reward-Related Behavior

Summary: This study reveals the important role of astrocyte circadian molecular clock function in the regulation of the nucleus accumbens (NAc) and reward-related behaviors. Approximately 43% of the astrocyte transcriptome shows diurnal rhythm, with key metabolic pathways enriched among the rhythmic genes. Disruption of molecular clock function in NAc astrocytes leads to increased locomotor response to novelty, exploratory drive, operant food self-administration, and motivation. These animals also exhibit disrupted metabolic gene expression, as well as altered glutamatergic signaling and gene expression in the NAc.

Astrocytes in the ventral pallidum extinguish heroin seeking through GAT-3 upregulation and morphological plasticity at D1-MSN terminals

Summary: The study found that astrocytes in the ventral pallidum exhibit increased expression of the GABA transporter GAT-3 after extinction of heroin seeking, which was not observed in animals undergoing heroin withdrawal without extinction training or after extinguishing sucrose seeking. Additionally, there was a selective increase in astrocyte proximity to dopamine D1-expressing terminals during extinction of heroin self-administration, suggesting the involvement of astrocyte-neuron interactions in behaviors associated with opioid use disorder.
