RNA uridyl transferases TUT4/7 differentially regulate miRNA variants depending on the cancer cell type

Ragini Medhi et al.

Summary: The human terminal uridyl transferases TUT4 and TUT7 play an important role in RNA regulation in cancer cells. Mutations in TUT4/7 lead to reduced miRNA uridylation and defects in cancer cell properties, as well as dysregulation of miRNA-mRNA networks in a cell-type-specific manner.

The dual role of glioma exosomal microRNAs: glioma eliminates tumor suppressor miR-1298-5p via exosomes to promote immunosuppressive effects of MDSCs

Yanhua Qi et al.

Summary: Clear evidence shows that tumors can use exosomes to secrete microRNAs for modulating the tumor microenvironment. In this study, miRNAs were found to be divided into exosome-enriched miRNAs and intracellular-retained miRNAs, with exosome-enriched miRNAs playing a dual role. Specifically, miR-1298-5p was enriched in CSF exosomes and demonstrated to suppress glioma progression while promoting the immunosuppressive effects of myeloid-derived suppressor cells (MDSCs). Mechanically, miR-1298-5p targeted SETD7 and MSH2 in glioma cells and MDSCs, respectively. These findings provide insights into the selective sorting of miRNAs into exosomes and their role in modulating tumor immunity in glioma.

RNA uridyl transferases TUT4/7 differentially regulate miRNA variants depending on the cancer cell type

Ragini Medhi et al.

Summary: In this study, the authors investigated TUT4/7-mediated RNA regulation in two cancer cell lines and found that mutations in TUT4/7 led to a decrease in miRNA uridylation, affecting cancer cell proliferation and migration. Unlike let-7 family miRNAs, most miRNAs do not require LIN28A for TUT4/7 regulation.

Glucose starvation suppresses gastric cancer through targeting miR-216a-5p/Farnesyl-Diphosphate Farnesyltransferase 1 axis

Ruiyang Zhao et al.

Summary: This study revealed that FDFT1 is downregulated in gastric cancer and is negatively correlated with disease severity. Overexpression of FDFT1 is associated with better prognosis in gastric cancer patients. miR-216a-5p was identified as a critical suppressor of FDFT1 expression, and targeting the miR-216a-5p/FDFT1 axis suppressed malignant behaviors of gastric cancer both in vitro and in vivo.

Cancer-secreted exosomal miR-1468-5p promotes tumor immune escape via the immunosuppressive of vessels

Chenfei Zhou et al.

Summary: The study suggests that miR-1468-5p in cervical cancer exosomes promotes lymphatic PD-L1 upregulation and lymphangiogenesis, leading to T cell immunity suppression and allowing cancer cells to evade anti-cancer immunity. High levels of exosomal miR-1468-5p in serum correlate with immunosuppressive status in the tumor microenvironment and poor prognosis in cervical cancer patients.

Impaired AGO2/miR-185-3p/NRP1 axis promotes colorectal cancer metastasis

Xisheng Liu et al.

Summary: The global downregulation of miRNA expression, potentially due to defects in miRNA processing machinery, is a hallmark of human cancer. In colorectal cancer (CRC), downregulation of the key regulator of miRNA processing, Argonaute 2 (AGO2), has been found to promote epithelial-mesenchymal transition (EMT) and indicate poor prognosis. Knockdown of AGO2 in CRC cells enhanced migration, invasion, and metastasis, while integrated analyses revealed neuropilin 1 (NRP1) as a new substrate of AGO2 via miR-185-3p. Therapeutically targeting the AGO2/miR-185-3p/NRP1 axis may offer a potential treatment approach for CRC.

Caveolin-1-driven membrane remodelling regulates hnRNPK-mediated exosomal microRNA sorting in cancer

Harley Robinson et al.

Summary: The study reveals a novel mechanism in which non-caveolar CAV1 drives the loading of metastasis-promoting microRNAs into exosomes by regulating hnRNPK localization, with this process being modulated by the lipid environment of membranes. Knockdown of hnRNPK in prostate cancer cells impairs the ability of extracellular vesicles to induce osteoclastogenesis, indicating a potential role in bone metastasis. Additionally, elevated levels of hnRNPK in biofluid extracellular vesicles are associated with metastatic prostate and colorectal cancer.
