Neurotoxicity of phenylalanine on human iPSC-derived cerebral organoids

Jieun Kim et al.

Summary: Phenylketonuria (PKU) is a genetic metabolic disorder that leads to the accumulation of phenylalanine in the blood. This study utilized human induced pluripotent stem cell (iPSC)-derived cerebral organoids to investigate the neurotoxicity of hyperphenylalaninemia. The findings suggest that phenylalanine exposure may contribute to microcephaly and abnormal cortical expansion in the developing human brain.

Thymidylate synthase drives the phenotypes of epithelial-to-mesenchymal transition in non-small cell lung cancer

Mohammad Aarif Siddiqui et al.

Summary: The study identified a functional correlation between thymidylate synthase (TS), EMT, chemotherapy and metastasis in non-small cell lung cancer (NSCLC), establishing TS as a potential therapeutic marker. Further experiments revealed that TS may influence the occurrence and development of NSCLC by regulating EMT transcription factors and effectors.

Methylenetetrahydrofolate (MTHFR), the One-Carbon Cycle, and Cardiovascular Risks

Shanel Raghubeer et al.

Summary: The MTHFR enzyme plays a crucial role in cellular homeostasis and its polymorphisms, especially C677T, are associated with various diseases including cardiovascular diseases. The C677T polymorphism leads to elevated Hcy levels and is considered an independent risk factor for CVD.

Metabolomic and transcriptomic analysis reveals endogenous substrates and metabolic adaptation in rats lacking Abcg2 and Abcb1a transporters

Samit Ganguly et al.

Summary: This study identified the potential roles of Bcrp and Pgp in endogenous substrate metabolism and central nervous system health through metabolomics and transcriptomics analysis. Significant changes in metabolite levels in dKO rats suggest that these transporters may impact multiple metabolic pathways.

Modulation of Urate Transport by Drugs

Summary: The interaction with the main reabsorptive urate transporter URAT1 seems to be dominant in determining the net effect of a drug on SU levels. In vitro transporter interaction studies can predict the effects of drugs on SU levels.
