The NADPARK study: A randomized phase I trial of nicotinamide riboside supplementation in Parkinson's disease

Brage Brakedal et al.

Summary: In this study, we found that oral intake of nicotinamide riboside (NR) in Parkinson's disease (PD) patients was safe and increased cerebral NAD levels, leading to altered cerebral metabolism and mild clinical improvement. NR also upregulated processes related to mitochondrial, lysosomal, and proteasomal function in blood cells and/or skeletal muscle, and decreased the levels of inflammatory cytokines.

Sirtuin 3 Dependent and Independent Effects of NAD+ to Suppress Vascular Inflammation and Improve Endothelial Function in Mice

Xiaoyun Cao et al.

Summary: Atherosclerosis is linked to endothelial dysfunction and vascular inflammation, and SIRT3 and NAD(+) play critical roles in maintaining normal mitochondrial function and improving endothelial function. The deletion of SIRT3 in endothelial cells accelerates atherosclerosis and amplifies vascular inflammation and oxidative stress. Treatment with NAD(+) attenuates these effects. The study also shows that NAD(+) supplementation improves endothelial function and reduces plaque formation in mouse models of atherosclerosis.

Moderate offspring exercise offsets the harmful effects of maternal protein deprivation on mitochondrial function and oxidative balance by modulating sirtuins

Anderson Apolonio Silva Pedroza et al.

Summary: Moderate intensity exercise during early life can mitigate the negative impact of maternal protein deprivation on cardiac bioenergetics by improving mitochondrial function and reducing oxidative stress. Additionally, exercise can counteract the deleterious effects of protein restriction by decreasing acetylation levels.

NAD+ boosting reduces age-associated amyloidosis and restores mitochondrial homeostasis in muscle

Mario Romani et al.

Summary: The study demonstrates dysregulation of mitochondrial and proteostasis pathways in muscle aging and diseases. Accumulation of amyloid-like deposits and mitochondrial dysfunction during natural aging can be partially ameliorated by boosting NAD(+) levels, highlighting a potential therapeutic approach for age-related muscle decline. Further clinical studies are warranted to explore the impact of NAD(+) metabolism-enhancing approaches on mitochondrial function and muscle homeostasis in aging.

Impact of gestational low-protein intake on embryonic kidney microRNA expression and in nephron progenitor cells of the male fetus

Leticia de Barros Sene et al.

Summary: This study found that gestational low-protein intake can affect renal development in offspring by altering miRNA and predicted target gene expression levels in the kidney, leading to a reduction in nephron number. Specifically, there were decreased cell numbers and changes in immunoreactivity of related proteins observed in the metanephros.

Post-translational modifications by SIRT3 de-2-hydroxyisobutyrylase activity regulate glycolysis and enable nephrogenesis

Luca Perico et al.

Summary: The study reveals a novel role for the NAD(+)-dependent deacylase SIRT3 in kidney development, controlling early renal development through the regulation of epigenetics and metabolic processes. Deficiency of SIRT3 leads to decreased nephron progenitors, impaired nephrogenesis, and a reduced number of nephrons in the developing kidney.

Opa1 relies on cristae preservation and ATP synthase to curtail reactive oxygen species accumulation in mitochondria

Ruben Quintana-Cabrera et al.

Summary: The study shows that the mitochondrial fusion and cristae shape protein Opa1 requires mitochondrial ATP synthase oligomers to reduce ROS accumulation. In some cells fueled with galactose to force ATP production by mitochondria, cristae are enlarged, ATP synthase oligomers destabilized, and ROS accumulate.

Podocyte endowment and the impact of adult body size on kidney health

Luise A. Cullen-McEwen et al.

Summary: Low birth weight due to poor maternal nutrition is associated with low podocyte endowment. A mild podocyte deficit at birth does not necessarily result in glomerular pathology in adulthood, but postnatal podocyte loss combined with excessive body weight can lead to albuminuria and glomerulosclerosis. These findings offer new insights into the relationships between birth weight, podocyte indexes, postnatal weight, and glomerular pathology.
