Noncanonical Metabotropic Glutamate Receptor 5 Signaling in Alzheimer's Disease

Summary: mGluR5 is widely expressed in the brain and plays a key role in memory and learning, synaptic transmission, and plasticity. Its dysfunction, specifically in response to Aβ42 oligomers, is believed to contribute to the pathophysiology of AD. As a potential therapeutic target for AD, recent studies have demonstrated the efficacy of mGluR5 allosteric modulators in improving memory deficits and mitigating disease pathology. However, the pharmacological differences and downstream signaling activation of mGluR5 in different genders suggest the need for reevaluation of its therapeutic potential in female AD patients.

Summary: Synaptic plasticity is crucial for learning and memory, and impaired plasticity can lead to memory deficits. Stimulation of the basolateral amygdala post-training can improve spatial learning and memory in lesioned rats by influencing the expression of plasticity-related genes in memory-associated brain regions. Both trained and BLA-stimulated lesioned rats showed significantly improved LTP in the dentate gyrus compared to non-trained lesioned rats, indicating a potential mechanism for ameliorating memory deficits.

Summary: Soluble amyloid-beta-protein (A beta) oligomers, a major hallmark of AD, trigger the integrated stress response (ISR) via multiple pathologies including neuronal hyperactivation, microvascular hypoxia, and neuroinflammation. Facilitation of mGluR5-LTD by A beta can be prevented by suppressing eIF2 alpha phosphorylation cascade with ISRIB, which also improves spatial learning and memory deficits caused by exogenous synthetic A beta-injected rats.

Changes in mGlu5 Receptor Signaling Are Associated with Associative Learning and Memory Extinction in Mice

Summary: This study investigates the effects of spatial learning and memory extinction on mGlu5 metabotropic glutamate receptor signaling in the hippocampus and prefrontal cortex. The results show that mGlu5 receptor-mediated PI hydrolysis is reduced in the dorsal hippocampus of learner mice compared to naive and control mice. Memory extinction is associated with an up-regulation of mGlu5 receptor-mediated PI hydrolysis in the three brain regions and an increase in mGlu5 receptor and phospholipase-C beta protein levels in the ventral and dorsal hippocampus, respectively.
