Recent advances in understanding the genetic basis of systemic lupus erythematosus

Eunji Ha et al.

Summary: Systemic lupus erythematosus (SLE) is a polygenic chronic autoimmune disease with a high heritability of up to 66%, with most risk variants residing in non-coding regions. Recent studies have identified dozens of new SLE loci and integrated GWAS signals with other biological resources in genetic associations. The research also explores genetic risk scores for SLE and their associations with clinical features, as well as variant-driven pathogenetic functions and future challenges in genetic studies on SLE.

Friend or foe: RIG- I like receptors and diseases

Jie Song et al.

Summary: This review provides a basic understanding of RLR biology, summarizes recent findings on the regulatory mechanisms of RLR signaling cascade, and focuses on the relationship between RLRs and diseases such as SARS-CoV-2, malaria, bacterial infections, autoimmune diseases, and anti-tumor immunity. These findings offer insights and guidance for antiviral and immunomodulatory therapies targeting RLRs.

A Novel Type I Interferon Primed Dendritic Cell Subpopulation in TREX1 Mutant Chilblain Lupus Patients

Anne Eugster et al.

Summary: This study analyzed the composition of dendritic cell subsets in patients with TREX1 deficiency and identified a novel myeloid dendritic cell subpopulation that is highly enriched in these patients and associated with upregulation of interferon pathway genes.

Molecular pathways in patients with systemic lupus erythematosus revealed by gene-centred DNA sequencing

Johanna K. Sandling et al.

Summary: The study identified two main independent pathways involved in SLE susceptibility: T lymphocyte differentiation and innate immunity, characterized by HLA and interferon, respectively. Pathway PRS could define pathways in individual patients, who on average were positive for seven pathways. Pathway PRS-based clustering allowed stratification of patients into four groups with different risk score profiles.

Abnormalities of the type I interferon signaling pathway in lupus autoimmunity

Stefania Gallucci et al.

Summary: Type I interferons (IFNs) play a crucial role in the pathogenesis of Systemic Lupus Erythematosus (SLE), and aberrant IFN signaling can exacerbate SLE. Genetic and epigenetic abnormalities are present in SLE patients. Neutralizing anti-type I IFN receptor antibodies may be a therapeutic strategy.

DsRNA Sequencing for RNA Virus Surveillance Using Human Clinical Samples

Takuma Izumi et al.

Summary: A new method for efficient detection of RNA viruses in human clinical samples was established, which showed higher viral read ratio compared to conventional methods and correctly identified viral RNA. This new technique will be an effective tool for intracellular RNA virus surveillance in human clinical samples and may be useful for the detection of new RNA viruses associated with diseases.

Human SLC15A4 is crucial for TLR-mediated type I interferon production and mitochondrial integrity

Toshihiko Kobayashi et al.

Summary: SLC15A4 is a crucial amino acid transporter that plays a significant role in regulating inflammatory responses and autophagy, genetically associated with inflammatory diseases like SLE and colitis. Research has shown that SLC15A4 plays a critical role in mediating inflammatory signaling through TLR7/8/9 and is important for maintaining autophagy processes.
