Circulating tumor cell gene expression and plasma AR gene copy number as biomarkers for castration-resistant prostate cancer patients treated with cabazitaxel

Giorgia Gurioli et al.

Summary: This study evaluated the prognostic role of CTC gene expression in cabazitaxel-treated mCRPC patients and its association with plasma AR copy number. The results showed that CTC gene expression was significantly associated with overall survival and disease progression. AR-V7 was identified as an important prognostic biomarker, and patients with AR-V7 positivity had poorer outcomes with lower dose cabazitaxel treatment.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Vertebrate Wnt5a-At the crossroads of cellular signalling

Sally Rogers et al.

Summary: Wnt signalling plays a crucial role in embryogenesis, cell motility, and tissue re-arrangement and organ formation in vertebrates. Wnt5a, a ligand of the Wnt signalling pathway, is involved in cell polarity and migration and has a broader function, including the regulation of other signalling pathways and crosstalk with the Wnt/beta-catenin pathway.

Improving the diagnosis of prostate cancer by telomerase-positive circulating tumor cells: A prospective pilot study

Zhenrong Yang et al.

Summary: This study investigated the diagnostic capability of circulating tumor cells (CTCs) detected by telomerase-based CTC detection (TBCD) in men with a PSA level of 4 ng/ml or higher. The results showed that CTCs had better diagnostic ability than PSA, especially in the range of 4-10 ng/ml. Additionally, a CTC+PSA combined model further improved the diagnosis of PSA levels in the 4-10 ng/ml range.

Automated enumeration and phenotypic characterization of CTCs and tdEVs in patients with metastatic castration resistant prostate cancer

Steffi Oeyen et al.

Summary: Automated CTC enumeration is reliable for prognosis assessment in mCRPC patients, and studying CTC phenotypic diversity adds value to patient prognostication beyond enumeration. High CTC phenotypic diversity before treatment is associated with shorter progression-free survival and overall survival, highlighting the importance of considering this factor in patient care.

Summary: This study investigated the associations of CTCs and CTC-clusters with mCRPC prognosis using longitudinal samples, finding that CTC counts were associated with mCRPC outcomes, and the presence of CTC-clusters alone had prognostic value, improving CTC-based prognostic stratification. The findings suggest the potential of combining CTC and CTC-clusters as non-invasive means to monitor progression and predict survival in mCRPC.

Platelets, immune cells and the coagulation cascade; friend or foe of the circulating tumour cell?

Mark P. Ward et al.

Summary: Circulating tumour cells (CTCs) are cancer cells that have unique characteristics allowing them to survive in the circulation, extravasate, and metastasize. Their interactions with components of the blood circulatory system influence their physical characteristics and metastatic capabilities, providing critical diagnostic and prognostic value in clinical settings. Platelets play a crucial role in the formation of metastatic niches and interact with CTCs to influence their fate, demonstrating potential for therapeutic disruption and targeted treatments in advanced cancers.

TRAIL-coated leukocytes to kill circulating tumor cells in the flowing blood from prostate cancer patients

Nerymar Ortiz-Otero et al.

Summary: The study revealed that radical surgery for localized prostate cancer can lead to tumor cell mobilization into the bloodstream, with these cells returning to baseline levels within a few weeks. In patients experiencing cancer recurrence, levels of circulating tumor cells and cancer-associated fibroblasts gradually increase over time. An experimental TRAIL-based therapy was shown to induce apoptosis in tumor cells in blood samples ex vivo, suggesting a potential new adjuvant therapy in combination with surgery.

Prospective Evaluation of Clinical Outcomes Using a Multiplex Liquid Biopsy Targeting Diverse Resistance Mechanisms in Metastatic Prostate Cancer

Summary: This study utilized a novel liquid biopsy technology to analyze mRNA from circulating tumor cells, identifying mechanisms of resistance to androgen receptor signaling inhibitors in prostate cancer. The expression of AR-Vs, AR genomic structural alterations, and neuroendocrine prostate cancer markers were found to play crucial roles in therapy optimization. The research demonstrated that a specific transcriptional profile in CTCs can serve as an independent prognostic marker, beyond AR-V7, in metastatic prostate cancer patients, helping to identify various ARSI resistance mechanisms.

Application of Primary/Secondary Circulating Tumor Cells for the Prediction of Biochemical Recurrence in Nonmetastatic Prostate Cancer Patients following Radical Prostatectomy or Radiotherapy: A Meta-Analysis

Liang Cao et al.

Summary: Our research suggests that the presence of secondary circulating tumor cells (SCTCs) is associated with a higher rate of biochemical recurrence (BCR) in patients with nonmetastatic prostate cancer (NMPC) after radical prostatectomy (RP) or radiotherapy (RT), while the presence of primary circulating tumor cells (PCTCs) does not show a significant association with higher BCR. It is recommended that NMPC patients undergo intensive CTC surveillance after RP or RT.
