Proteomic Profiling and Biomarker Discovery in Colorectal Liver Metastases

Geoffrey Yuet Mun Wong et al.

Summary: Colorectal liver metastases (CRLM) are a major cause of death in patients with metastatic colorectal cancer (CRC). Liver resection is the main treatment for select patients with CRLM, but effective treatment remains challenging due to high rates of recurrence. Genomic profiling has improved our understanding of CRC, but the functional context at the protein level is not well-established. Proteomic biomarkers are being explored for predicting CRLM and recurrence. This review aims to rationalize the proteomic complexity of CRC and explore the potential applications of proteomic biomarkers in CRLM.

The effects of resection margin and KRAS status on outcomes after resection of colorectal liver metastases

Ayiesha Ahmad Zalmani Hatta et al.

Summary: The study found that in patients with resected CRLM, those with mt-KRAS mutations had significantly shorter overall survival compared to those with wt-KRAS. Additionally, smaller margins were associated with shorter recurrence-free survival in wt-KRAS patients, but not in mt-KRAS patients. This information could be valuable for surgical planning and post-operative counseling based on margin and KRAS status.

Preliminary Analysis of Liquid Biopsy after Hepatectomy for Colorectal Liver Metastases

Meredith C. Mason et al.

Summary: Liquid biopsies are increasingly used in patients with colorectal cancer to evaluate tumor burden, treatment response, and prognosis. After resection of colorectal liver metastases (CLMs), positive liquid biopsy results are associated with worse overall survival, particularly when multiple gene mutations are detected. Liquid biopsy provides complementary information about disease burden and prognosis compared to serum CEA and CT imaging.

Summary: This study observed significant hospital variation in the Netherlands for combined resection and ablation procedures. Short-term postoperative outcomes, including hospital stay, postoperative complicated course, and 30-day mortality, were better in the resection and ablation group compared to resection alone group, attributed to avoiding major hepatectomy-related complications. However, these differences faded in multivariable logistic regression analysis.
