Host Genetic Associations with Salivary Microbiome in Oral Cancer

S. F. Yang et al.

Summary: This study conducted a microbiome genome-wide association study in patients with oral squamous cell carcinoma and noncancer individuals, identifying associations between specific genetic loci and oral microbiota composition. These loci were found to be enriched for genes involved in the regulation of oncogenic and angiogenic responses, highlighting a genetic influence on the structure of oral microbial communities.

A dysbiotic microbiome promotes head and neck squamous cell carcinoma

Daniel N. Frank et al.

Summary: Recent studies have found dysbiotic oral microbiota and tumor-resident bacteria in patients with HNSCC, with elevated levels of Lactobacillus spp. and reduced levels of Neisseria spp. Furthermore, experimental evidence shows that depletion of microbiota delays oral tumorigenesis, while transfer of microbiota from mice with oral cancer accelerates tumorigenesis. These findings support the hypothesis that dysbiosis promotes HNSCC development.

Dysbiotic tumor microbiota associates with head and neck squamous cell carcinoma outcomes

Yu Dou et al.

Summary: The study reveals an association between tumor microbiota and outcomes of HNSCC patients, with poor prognosis patients having lower microbial richness and specific genera enrichment, while good prognosis patients having richer and health-promoting microbial communities. The microbial dysbiosis index (MDI) shows significant association with overall survival (OS) and disease free survival (DFS), independently of other clinical factors.

Candida albicans Enhances the Progression of Oral Squamous Cell Carcinoma In Vitro and In Vivo

Mate Vadovics et al.

Summary: Oral Candida albicans infection is associated with the development and progression of oral squamous cell carcinoma (OSCC). Candida albicans promotes the progression of OSCC by stimulating the production of matrix metalloproteinases, oncometabolites, protumor signaling pathways, and overexpression of prognostic marker genes associated with metastatic events. Candida albicans also upregulates oncogenes in nonmalignant cells.

Metagenomic Analysis Reveals a Changing Microbiome Associated With the Depth of Invasion of Oral Squamous Cell Carcinoma

Yuan Liu et al.

Summary: The relationship between oral squamous cell carcinoma (OSCC) development and the oral microbiome has been studied using metagenomic analysis. The abundance of certain bacteria is associated with the stage of the disease and has predictive value for different stages of OSCC. In addition, the functional analysis showed changes in metabolic pathways, flagellar assembly, bacterial chemotaxis, and other factors as the disease progressed. These findings provide insights into the role of the microbiome in OSCC and its potential for diagnosis and treatment.

Biomaterial-mediated modulation of oral microbiota synergizes with PD-1 blockade in mice with oral squamous cell carcinoma

Di-Wei Zheng et al.

Summary: It has been demonstrated that modulating the microbiota in tumors can enhance antitumor immune responses and improve cancer patient survival rates. The use of hydrogel-embedded silver nanoparticles and specific exogenous bacteria shows potential in boosting the effectiveness of cancer therapies by influencing the host's immune system through host-microbiota interactions.

Comparative and analytical characterization of the oral bacteriome of smokeless tobacco users with oral squamous cell carcinoma

Ankita Srivastava et al.

Summary: This study investigated the oral bacteriome of smokeless tobacco users with oral cavity squamous cell carcinoma (OSCC) and found significant differences compared to healthy smokeless tobacco users and non-users. The prevalence of tobacco-specific nitrosamines forming bacteria was observed in the patients with OSCC. The functional attributes of the oral bacteriome were correlated with genes involved in oncogenesis.

Emerging role of microbiota in immunomodulation and cancer immunotherapy

Raghwendra Pratap Singh et al.

Summary: The gut microbiota plays a crucial role in modulating the immune system and impacting the pathophysiology of diseases like cancer. Research suggests that the gut microbiota can influence both carcinogenesis and the efficacy of cancer therapy, including immunotherapy. Understanding the mechanisms behind microbiota-mediated immune response modulation is essential for utilizing them as cancer therapeutics, potentially transforming current treatment strategies into personalized cancer therapies.

Summary: Chemoradiation can lead to oral dysbiosis with depletion of specific genera crucial for the enterosalivary nitrate-nitrite-nitric oxide pathway. The oral metabolomic profiles also show a significant downregulation of nitric oxide-related compounds in saliva samples from patients postchemoradiation, indicating a potential impact on quality of life for OCC and OPC patients post-treatment.
