A targeted nanoplatform co-delivery of pooled siRNA and doxorubicin for reversing of multidrug resistance in breast cancer

Summary: This study aimed to develop a novel and facile strategy for preparing a highly efficient and safe gene and drug delivery system for multidrug-resistant (MDR) breast cancer. A targeted nanoplatform based on biodegradable boronic acid modified epsilon-polylysine was developed to co-deliver P-gp siRNA, Bcl-2 siRNA, and doxorubicin for overcoming MDR. The nanoplatform exhibited robust inhibitory effects against invasion, proliferation, and colony formation of MCF-7/ADR cells in vitro, and demonstrated ATP responsiveness. In vivo antitumor experiments showed significant inhibition of tumor growth and good biocompatibility with the targeted nanoplatform.

Summary: This study demonstrates a self-assembled strategy for simultaneous delivery of the therapeutic drug salinomycin (SAL) and siRNA to cancer cells using chitosan nanoparticles. The siRNA@C-SAL showed superior cytotoxicity against gastric carcinoma cells without significant adverse effects in healthy mice. Thus, this improved drug delivery system with siRNA and hydrophobic drugs provides a potential strategy for the treatment of gastric cancer.

Combining parthenolide (PTL) and cisplatin (DDP) has been shown to synergistically inhibit NSCLC growth by affecting the PI3K-AKT signaling pathway. This combination therapy may offer a new option for treating NSCLC.

Cisplatin and si-Notch1-Folic Acid-Conjugated Mesoporous Silica Nanoparticles Prevent Hepatocellular Carcinoma
