Who cares about oligozoospermia when we have ICSI

Sandro C. Esteve

Summary: The value of assessing subfertile males with oligozoospermia is controversial, but offering indiscriminate ICSI is not the best choice. Oligozoospermia is associated with poor male reproductive health, sperm damage, and adverse health consequences for offspring. Many testicular conditions can cause oligozoospermia, and therapeutic interventions can improve sperm quality and male health. Fertility clinics should collaborate with male infertility specialists to diagnose and treat oligozoospermia for better patient health, pregnancy prospects, and offspring well-being.

Elevated Expression of lncRNA MEG3 Induces Endothelial Dysfunction on HUVECs of IVF Born Offspring via Epigenetic Regulation

Ying Jiang et al.

Summary: Cardiovascular dysfunction in children born after in vitro fertilization (IVF) has been a concern, but the underlying molecular mechanisms are still unknown. This study found that IVF born offspring had slightly higher systolic blood pressure at 2 years old compared to naturally conceived children. The expression of maternally expressed gene 3 (MEG3) was higher in IVF offspring's umbilical vein endothelial cells (HUVECs) and correlated with their blood pressure. Additionally, IVF offspring exhibited decreased expression of endothelial nitric oxide synthase (eNOS) and vascular endothelial growth factor (VEGF), along with increased expression of endothelin-1 (ET1).

DNA methylation profiles after ART during human lifespan: a systematic review and meta-analysis

Summary: ART procedures might influence DNA methylation in fetus and placenta, potentially leading to increased risks of adverse outcomes associated with ART. Despite the growing body of literature on this topic, the conclusions from existing studies remain inconsistent, and the safety of ART at the epigenetic level is still uncertain.

Contemporary Use of ICSI and Epigenetic Risks to Future Generations

Summary: This review discusses the use of intracytoplasmic sperm injection (ICSI) in assisted reproductive technology (ART) and the potential risks associated with it. While ICSI has been successful in helping individuals with infertility, there is growing evidence that it may lead to adverse health outcomes in offspring, including epigenetic disorders and other abnormalities.

OS has detrimental effects on gametes and embryos, necessitating strategies to ameliorate its impact during the entire embryo culture process.

Do assisted reproductive technologies and in vitro embryo culture influence the epigenetic control of imprinted genes and transposable elements in children?

J. Barberet et al.

Summary: This study compared the epigenetic profiles of children conceived through ART and naturally conceived children, finding significant differences in DNA methylation levels of IGs between the two groups. While there were no differences found in the culture media, specific IG regions related to development showed methylation changes between children conceived through ART and naturally conceived children.

Among offspring conceived by ART, multiple pregnancies may lead to a higher risk of urogenital tract malformations compared to singleton pregnancies.

DNA methylation patterns within whole blood of adolescents born from assisted reproductive technology are not different from adolescents born from natural conception

B. Penova-Veselinovic et al.

Summary: The DNA methylation profiles of adolescents born from ART (IVF or ICSI) were found to be not significantly different from their naturally conceived counterparts. This suggests that the effects of ART on DNA methylation are transient and do not persist into adolescence. However, alternative epigenetic mechanisms may still be at play.

Summary: This study conducted a meta-analysis on the correlation between assisted reproductive technology (ART) conception and the risk of autism spectrum disorder (ASD) in offspring, finding a positive correlation between ART and ASD while no significant association was observed in singletons. Future high-quality prospective studies with larger sample sizes are needed to further validate these findings.

Do frozen embryo transfers modify the epigenetic control of imprinted genes and transposable elements in newborns compared with fresh embryo transfers and natural conceptions?

Julie Barberet et al.

Summary: The study found that compared with natural conceptions and frozen embryo transfers, fresh embryo transfers were associated with changes in methylation and/or transcription of some TEs and IGs, mostly in placental samples, which could be indicative of altered placental epigenetic regulation resulting from ovarian stimulation protocols.

While some associations were detected in two study cohorts, they were not replicated in another cohort, suggesting the need for further research to identify replicable variation in DNA methylation due to medically assisted reproduction.

Relevance of sperm imprinted gene methylation on assisted reproductive technique outcomes and pregnancy loss: a systematic review

Rossella Cannarella et al.

Summary: Recent evidence suggests that altered methylation of sperm-imprinted genes, particularly H19/IGF2 and KCNQ1, may be associated with sperm DNA fragmentation (SDF), pregnancy loss, and assisted reproductive technique (ART) outcomes. However, the methylation status of other genes such as GLT2, PEG1/MEST, and ZAC/PLACL1 may not have significant impacts on these factors. Further research is needed to confirm these findings and explore the potential epigenetic predictors of ART outcomes.
