Missense variants in the N-terminal domain of the A isoform of FHF2/FGF13 cause an X-linked developmental and epileptic encephalopathy

Andrew E. Fry et al.

Summary: FHFs are intracellular proteins that regulate Na-v channels and their dysfunction has been linked to neurological disorders including epilepsy. Mutant FHF2A proteins were found to lose the ability to induce rapid-onset, long-term blockade of Na-v channel while retaining pro-excitatory properties, increasing neuronal excitability and potentially causing epileptic seizures.

A SCN8A variant associated with severe early onset epilepsy and developmental delay: Loss- or gain-of-function?

Yuanyuan Liu et al.

Summary: The study revealed that the variant p.(Arg223Gly) in the SCN8A gene leads to neuronal hyperexcitability and increased firing rate, consistent with the central role of gain-of-function (GOF) mechanisms in developmental and epileptic encephalopathy (DEE).

Early onset epilepsy and sudden unexpected death in epilepsy with cardiac arrhythmia in mice carrying the early infantile epileptic encephalopathy 47 gain-of-function FHF1(FGF12) missense mutation

Jana Veliskova et al.

Summary: The Arg52His mutation in the FHF1 gene leads to severe seizures and sudden death in mice, with abnormal heart rhythm suggesting a functional axis between FHF1 and Na(v)1.6 underlying changes in brain sodium channel gating in epileptic encephalopathy.
