Identification and Quantification of Necroptosis Landscape on Therapy and Prognosis in Kidney Renal Clear Cell Carcinoma

Summary: In this study, the necroptosis landscape of KIRC patients was quantified and two distinct necroptosis-related patterns were identified. A stable and accurate prognostic marker called NRGscore was constructed, which showed significant correlations with clinicopathological characteristics, tumor immune microenvironment, and tumor mutation burden of KIRC patients. NRGscore also had effective guiding significance for immunotherapy, chemotherapy, and targeted drugs.

Identification and verification of the ferroptosis- and pyroptosis-associated prognostic signature for low-grade glioma

Summary: Accumulating evidence suggests that ferroptosis and pyroptosis play important roles in the tumorigenesis of low-grade glioma (LGG). This research aimed to classify molecular subtypes of LGG and identify a novel multigene signature based on ferroptosis- and pyroptosis-related genes. The study found that the FPRGs had prognostic value and developed a six-gene prognostic signature that outperformed other recognized signatures in predicting the survival probability of LGG patients.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Summary: HIF1 alpha plays an important role in pan-cancer, with its expression levels correlating with tumor stage and prognosis. It is involved in multiple small molecule pathways. Head and neck cancer has the highest mutation frequency of the HIF1 alpha gene, and HIF1 alpha methylation level is reduced in most tumors. HIF1 alpha is not only associated with immune cell infiltration but also with immune checkpoint genes and tumor mutation burden.

Construction and Identification of a Novel 5-Gene Signature for Predicting the Prognosis in Breast Cancer

Summary: This study identified hub genes and constructed a prognostic signature for predicting survival of breast cancer patients. The 5-gene signature (GP6, MAK, DCTN2, TMEM156, and FKBP14) showed favorable prediction performance and was confirmed to be an independent risk indicator. High M2 macrophage scores and TP53 mutations were associated with poor prognosis in the high-risk group, while high CD8+ T cell scores and CDH1 mutations were associated with good prognosis in the low-risk group. Patients in the low-risk group also showed a good response to immunotherapy.

Comprehensive analysis of the cancer driver genes in breast cancer demonstrates their roles in cancer prognosis and tumor microenvironment

Summary: This study identified over 200 differentially expressed driver genes and 27 prognosis-related genes in breast cancer. Patients in the high-risk group had lower survival rates compared to the low-risk group (P<0.05), and the risk signature showed high specificity and sensitivity in predicting patient prognosis (AUC 0.790). Multivariate regression analysis indicated that risk scores can independently predict patient prognosis. Additionally, differences in PD-1 expression, immune score, and stromal score were found among different risk groups.
