m7G Methylation-Related Genes as Biomarkers for Predicting Overall Survival Outcomes for Hepatocellular Carcinoma

Xin-Yu Li et al.

Summary: The study aimed to search for prognostic biomarkers for hepatocellular carcinoma (HCC) and construct a prognostic risk model based on N7-methyladenosine (m7G) methylation regulators. The results showed that the developed model had good predictive power, and a high-risk score was found to be an independent risk factor for the prognosis of HCC patients.

Roles of RNA Methylation on Tumor Immunity and Clinical Implications

Maorun Zhang et al.

Summary: RNA methylation is a widely existing RNA modification in eukaryotes and prokaryotes, affecting various aspects of RNA such as splicing, nucleation, stability, and immunogenicity. It plays a role in many physiological and pathological processes, particularly in the immune system. Through regulating RNA immunogenicity and innate immune components, RNA methylation influences the innate immunity of the body.

Chromatin-state barriers enforce an irreversible mammalian cell fate decision

M. Andres Blanco et al.

Summary: The study investigates the balance between self-renewal and differentiation in stem and progenitor cells, as well as the point at which progenitors irreversibly lose the capacity for self-renewal and commit to differentiation. The findings demonstrate that irreversible commitment is primarily due to loss of open regulatory site access and disruption of a positive feedback transcription factor activation loop.

Highly immunogenic cancer cells require activation of the WNT pathway for immunological escape

Yoshiko Takeuchi et al.

Summary: Highly mutated NSCLC with activation of the WNT/β-catenin pathway leads to impaired immune cell infiltration into the tumor microenvironment, resulting in resistance to PD-1 blockade therapy. Combination therapy with PD-1 blockade and WNT/β-catenin signaling inhibitors shows enhanced antitumor immunity compared to monotherapy.

METTL1 promotes hepatocarcinogenesis via m7G tRNA modification-dependent translation control

Zhihang Chen et al.

Summary: The N-7-methylguanosine (m(7)G) modification is a common transfer RNA (tRNA) modification in humans. Elevated levels of METTL1 and WDR4 in hepatocellular carcinoma (HCC) are associated with advanced tumor stages and poor patient survival. Mechanistically, METTL1-mediated tRNA m(7)G modification promotes the translation of target mRNAs with higher frequencies of m(7)G-related codons, providing new insights into the role of misregulated tRNA modifications in liver cancer.

N7-Methylguanosine tRNA modification enhances oncogenic mRNA translation and promotes intrahepatic cholangiocarcinoma progression

Zihao Dai et al.

Summary: Intrahepatic cholangiocarcinoma (ICC) shows upregulation of N-7-methylguanosine (m(7)G) tRNA modification and its methyltransferase complex components, METTL1 and WDR4, which are associated with poor prognosis. METTL/WDR4 play a critical role in promoting ICC cell survival and progression by selectively regulating the translation of oncogenic transcripts through m(7)G-tRNA-decoded codon-frequency-dependent mechanisms. Studies with overexpression and knockout mouse models demonstrate the crucial oncogenic function of Mettl1-mediated m(7)G tRNA modification in promoting ICC tumorigenesis and progression in vivo.

m7G Methyltransferase METTL1 Promotes Post-ischemic Angiogenesis via Promoting VEGFA mRNA Translation

Yongchao Zhao et al.

Summary: This study identified METTL1 as responsible for a global decrease of m7G in mRNA from ischemic tissues, and gene transfer of METTL1 improved blood flow recovery and angiogenesis. In vitro experiments showed that increased METTL1 expression promoted HUVECs proliferation, migration, and tube formation. Mechanistically, METTL1 facilitated VEGFA mRNA translation in an m7G methylation-dependent manner.

N6-Methyladenosine RNA Methylation Regulator-Related Alternative Splicing (AS) Gene Signature Predicts Non-Small Cell Lung Cancer Prognosis

Zhenyu Zhao et al.

Summary: Aberrant N6-methyladenosine (m6A) RNA methylation regulatory genes and related gene alternative splicing (AS) were found to be predictors of non-small cell lung carcinoma prognosis. By analyzing the expression profiles and splicing events of m6A regulatory genes in LUAD and LUSC, a prognostic signature for AS events was constructed to predict the prognosis of both types of lung cancer, with different AS genes associated with different clinical features and patient outcomes.

Summary: Lung cancer, the leading cause of cancer-related mortality globally, involves genetic and epigenetic events in the respiratory epithelium. Epigenetic modifications, including DNA methylation and RNA modifications, play a crucial role in lung cancer development. Various RNA nucleotide modifications, such as m6A, have recently gained attention for their role in biological processes and pathology. Dysregulation of RNA modifications can contribute to carcinogenesis and other pathological processes, emphasizing the importance of understanding RNA epigenetics for potential therapeutic targets in pulmonary diseases.
