Synthesis and Evaluation of Bakuchiol Derivatives as Potent Anti-inflammatory Agents in Vitro and in Vivo

Summary: Bakuchiol derivative 7a showed stronger anti-inflammatory activity than bakuchiol and celecoxib. Mechanistic studies revealed that 7a exerted its anti-inflammatory effects by regulating the release of cytokines and activating specific signaling pathways. In vivo experiments in zebrafish confirmed the anti-inflammatory activity of 7a.

Design, synthesis and evaluation of dihydrotriazine derivatives-bearing 5-aryloxypyrazole moieties as antibacterial agents

Summary: A series of dihydrotriazine derivatives with 5-aryloxypyrazole moieties were synthesized and evaluated for their antibacterial and antifungal activity. Compound 10d showed the strongest activity with significant inhibition against various strains, potentially through DHFR inhibition. Additionally, the compound exhibited significant inhibition of biofilm formation in bacterial cells at its minimum inhibitory concentration.

Novel Cinnamic Acid Derivatives Containing the 1,3,4-Oxadiazole Moiety: Design, Synthesis, Antibacterial Activities, and Mechanisms

Summary: A cinnamic acid derivative containing the 1,3,4-oxadiazole moiety (compound 31) showed promising antibacterial activity against rice bacterial leaf streak, with a 50% effective concentration of 0.2 mg/L against Xanthomonas oryzae pv. oryzicola. The compound exhibited curative and protective activities against the disease at 100 mg/L, with values higher than the control treatment.

Synthesis and evaluation of mycophenolic acid derivatives as potential anti-Toxoplasma gondii agents

Summary: Nineteen mycophenolic acid derivatives were synthesized, with compound E5 showing the strongest anti-Toxoplasma activity. E5 demonstrated significant protective effects on the host during acute Toxoplasma infection, indicating its potential as a candidate anti-T. gondii drug. Additionally, E5 exhibited hepatoprotective effects against T. gondii infection and relieved hepatomegaly and splenomegaly induced by acute Toxoplasma infection.

Anti-cancer effect of metformin on the metastasis and invasion of primary breast cancer cells through mediating NF-kB activity

Summary: This in vitro study demonstrated that metformin can inhibit the proliferation and metastasis of primary breast cancer cells by modulating MMP-2 and MMP-9 expressions and interfering with the NF-kB signaling pathway. The highest inhibition was observed at a dose of 25 mM Metformin, which downregulated MMP-9 expression and blocked NF-kB activity and nuclear translocation. Metformin may play an essential antitumor role in the invasion and metastasis pathways of PBCCs.

One of the compounds showed strong antifungal activity with high selectivity for Candida albicans. This compound exhibited significantly higher activity against C. albicans compared to the positive control compounds.

Summary: Standard chemotherapy and personalized target therapies are commonly used in advanced nonsmall cell lung cancer patients, but multidrug resistance and tumor metastasis often lead to decreased efficacy, associated with epithelial-mesenchymal transition (EMT). The transcription factor Twist1 plays a key role in promoting EMT, MDR, and metastasis, making it a potential target for developing antitumor therapies. By designing novel benzo[d] imidazo[2,1-b]thiazole derivatives with 1,3,4-oxadiazole moiety based on the structure of Twist1 inhibitor harmine, a compound with superior anti-proliferative activity and Twist1 down-regulation potential was identified as a promising lead for antineoplastic treatment.
