A redox cycle with complex II prioritizes sulfide quinone oxidoreductase-dependent H2S oxidation

Roshan Kumar et al.

Summary: The dual roles of H2S as a respiratory substrate and toxin raise questions about how it is cleared when the electron transport chain is inhibited. This study discovered a newly established redox cycle between SQOR and complex II at high H2S concentrations, which plays a significant role in prioritizing H2S oxidation.

Metabolic determination of cell fate through selective inheritance of mitochondria

Julia Dohla et al.

Summary: The study demonstrates that selectively and asymmetrically inherited mitochondria in mammary stem-like cells impose a metabolic bias on progeny, affecting the balance between self-renewal and differentiation. This metabolic feature, established by functionally distinct mitochondrial age-classes, acts as a fate determinant in asymmetric division of epithelial stem-like cells.Cells that inherit old mitochondria with oxidative energy metabolism promote differentiation, while cells inheriting newly synthesized mitochondria with low levels of Rieske iron-sulfur polypeptide 1 maintain stemness through the pentose phosphate pathway during early fate determination.

Overproduction of hydrogen sulfide, generated by cystathionine β-synthase, disrupts brain wave patterns and contributes to neurobehavioral dysfunction in a rat model of down syndrome

Theodora Panagaki et al.

Summary: This study investigates the functional role of the cystathionine-beta-synthase (CBS)/hydrogen sulfide (H2S) pathway in the pathogenesis of brain wave pattern alterations and neurobehavioral dysfunction in a rat model of Down syndrome (DS). The overexpression of CBS and subsequent overproduction of H2S in the brain of DS rats were observed. The study suggests that the CBS/H2S pathway contributes to the neurological dysfunction in DS through dysregulation of cellular bioenergetics and gene expression.

Functional, metabolic and transcriptional maturation of human pancreatic islets derived from stem cells

Diego Balboa et al.

Summary: Transplanting pancreatic islet cells derived from human pluripotent stem cells has great potential for treating diabetes. This study generated functionally mature stem-cell-derived islets (SC-islets) using an optimized protocol and compared them comprehensively to primary adult islets. The results showed that SC-islets exhibited similar functional properties to adult islets in terms of insulin secretion, electrophysiology, signaling, and exocytosis. Additionally, single-cell transcriptomics revealed a continuous maturation trajectory of SC-islets in vitro and during engraftment in mice, closely resembling that of primary islets.

3-Mercaptopyruvate sulfurtransferase: an enzyme at the crossroads of sulfane sulfur trafficking

Brandan Pedre et al.

Summary: 3-Mercaptopyruvate sulfurtransferase (MPST) is involved in hydrogen sulfide generation, protein modification, and cyanide detoxification, while also playing a crucial role in oxidative stress resistance, mitochondrial function, and fatty acid metabolism. However, the overall role and regulation of MPST in the broader physiological context remain to be fully understood.

Hydrogen sulfide-induced GAPDH sulfhydration disrupts the CCAR2-SIRT1 interaction to initiate autophagy

Iram Khan Iqbal et al.

Summary: This study demonstrates that hydrogen sulfide sulfhydrates GAPDH, leading to its redistribution into the nucleus and subsequently activating SIRT1 and autophagy. This pathway plays a crucial role in restricting the growth of intracellular Mycobacterium tuberculosis through autophagy-mediated trafficking.

Pharmacological induction of mesenchymal-epithelial transition via inhibition of H2S biosynthesis and consequent suppression of ACLY activity in colon cancer cells

Kelly Ascencao et al.

Summary: Hydrogen sulfide (H2S) is an important endogenous gaseous transmitter mediator that regulates cellular functions. In cancer cells, overproduction of H2S has been linked to processes such as metabolism stimulation and cell proliferation. By inhibiting H2S biosynthesis in colon cancer cells, a shift towards MET was induced, potentially contributing to anti-cancer effects.

Profiling Basal Forebrain Cholinergic Neurons Reveals a Molecular Basis for Vulnerability Within the Ts65Dn Model of Down Syndrome and Alzheimer's Disease

Melissa J. Alldred et al.

Summary: The basal forebrain cholinergic neuron degeneration is a common feature in Down syndrome and Alzheimer's disease. Utilizing the Ts65Dn trisomic mouse model, researchers identified novel therapeutic targets by analyzing transcriptomic profiles of medial septal nucleus BFCNs. The dysregulated transcriptomic profile of trisomic BFCNs provides key information on selective vulnerability within the septohippocampal circuit, offering potential insights for therapeutic approaches in DS and AD.

Summary: Initially considered a toxic gas, further research has shown that H2S has physiological roles and can act as a signaling molecule, working in synergy with NO to relax vascular smooth muscle. H2Sn, produced by 3MST, regulate the activity of ion channels and enzymes, and even affect tumor growth.
