Autophagy enhanced by curcumin ameliorates inflammation in atherogenesis via the TFEB-P300-BRD4 axis

Xuesong Li et al.

Summary: Disturbance of macrophage-associated lipid metabolism plays a key role in atherosclerosis. The crosstalk between autophagy deficiency and inflammation response in foam cells (FCs) through epigenetic regulation is poorly understood. In this study, it was observed that oxidized low-density lipoprotein (ox-LDL) led to macrophage autophagy deficiency and abnormal crosstalk between autophagy and inflammation. The dysfunctional transcription factor EB (TFEB)-P300-bromodomain-containing protein 4 (BRD4) axis mediated aberrant lipid metabolism. ox-LDL activated BRD4-dependent super-enhancer associated with liquid-liquid phase separation (LLPS) on the regulatory regions of inflammatory genes. Curcumin (Cur) restored FCs autophagy by promoting TFEB nuclear translocation, optimizing lipid catabolism, and reducing inflammation. The anti-atherogenesis effect of Cur was inhibited by macrophage-specific Brd4 overexpression or Tfeb knock-out in Apoe knock-out mice via bone marrow transplantation. These findings establish a novel TFEB-P300-BRD4 axis and highlight the epigenetic regulatory mechanisms by which Cur regulates autophagy, inhibits inflammation, and decreases lipid content.

Summary: Inflammatory bowel diseases (IBDs) are systemic diseases that can affect not only the gut but also other organs, impacting the quality of life for patients. Understanding the prevalence, pathophysiology, and clinical presentation of extraintestinal manifestations (EIMs) is crucial for effective IBD treatment. EIMs commonly affect joints, skin, or eyes, but may also involve other organs.

Curcumin analogue C66 attenuates obesity-induced renal injury by inhibiting chronic inflammation

Lin Ye et al.

Summary: Obesity is a major risk factor for chronic kidney disease, but the curcumin derivative C66 has been found to attenuate obesity-induced renal injury by inhibiting chronic inflammation and apoptosis through targeting NF-κB and JNK pathways, suggesting its potential in preventing obesity-associated renal diseases.
