Enhancing therapeutic performance of personalized cancer vaccine via delivery vectors

Tong Ye et al.

Summary: Personalized cancer vaccines have gained attention for their ability to overcome interindividual differences and show benefits for individual patients in the clinic. Various delivery vectors, including artificial materials, engineered microorganisms, cells and cell derivatives, have been studied to enhance the therapeutic performance of personalized cancer vaccines. These delivery vectors are utilized to improve antigen distribution and immune response strength, velocity, and duration through different delivery strategies.

The Broad Immunomodulatory Effects of IL-7 and Its Application In Vaccines

Juan Huang et al.

Summary: IL-7 is a crucial cytokine produced by various cells in the body, playing a key role in regulating lymphocyte development, survival, and immune responses against pathogens. Its functions include orchestrating gene rearrangement in precursor B and T cells, activating immune cells, and improving vaccine efficacy by enhancing immune responses.

Recombinant Mycobacterium smegmatis delivering a fusion protein of human macrophage migration inhibitory factor (MIF) and IL-7 exerts an anticancer effect by inducing an immune response against MIF in a tumor-bearing mouse model

Hyein Jeong et al.

Summary: The rSmeg-hMIF-hIL-7 vaccine demonstrated strong anti-tumor immune response in mice by inhibiting MIF-dependent tumorigenesis promotion and enhancing cytotoxic T cell recruitment into tumor microenvironments. It also showed enhanced anticancer effect with immune checkpoint inhibitors, suggesting its potential as an adjuvant for cancer immunotherapy. This study is the first to demonstrate the anticancer potential of an immunotherapeutic vaccine targeting immune response against MIF.

IL-7 expands lymphocyte populations and enhances immune responses to sipuleucel-T in patients with metastatic castration-resistant prostate cancer (mCRPC)

Russell K. Pachynski et al.

Summary: Treatment with rhIL-7 in patients with mCRPC after sip-T led to significant expansion of CD4+ and CD8+ T cells, as well as CD56(bright) natural killer cells, along with improved antigen-specific humoral and T cell proliferative responses over time. Additionally, analysis revealed increased expression of activation markers and beneficial cytokines.

Therapeutic cancer vaccines

Mansi Saxena et al.

Summary: Therapeutic cancer vaccines aim to induce tumor regression, eradicate minimal residual disease, establish lasting anti-tumor memory, and avoid non-specific or adverse reactions. However, challenges arise due to tumor-induced immunosuppression and immunoresistance, hindering the achievement of these goals.

Cancer immunotherapy with T-cell targeting cytokines: IL-2 and IL-7

Ji-Hae Kim et al.

Summary: This review discusses the positive correlation between increased effector cells, especially tumor-specific T cells, and patients' prognosis in clinical trials. It also highlights the limitations for patients with poor prognosis due to the lack of either T cells or targets for checkpoint inhibitors (CPIs). Furthermore, the focus is on the basic biology of how interleukin (IL)-2 and IL-7 regulate T-cell response and their clinical trials in cancer treatment, as well as recent studies aiming to improve cytokines' biological activities and identify strategies for combination therapy.
