Humoral and cellular immune memory to four COVID-19 vaccines

Zeli Zhang et al.

Summary: Multiple COVID-19 vaccines have successfully protected against symptomatic cases and deaths. Comparisons of T cell, B cell, and antibody responses to different vaccines can provide insights into protective immunity against COVID-19, particularly immune memory. mRNA vaccines and Ad26.COV2.S induced strong T cell responses, while mRNA vaccines showed substantial declines in antibodies.

Mapping the developing human immune system across organs

Chenqu Suo et al.

Summary: This study used single-cell genomics to decode the development of the human immune system during prenatal stages, revealing new cell types and the timing of immune function acquisition. The findings have important implications for cell engineering, regenerative medicine, and disease understanding.

Single-nucleus cross-tissue molecular reference maps toward understanding disease gene function

Gokcen Eraslan et al.

Summary: Understanding the function and regulation of genes in homeostasis and disease requires knowledge of their expression in different cellular and tissue contexts. In this study, we performed single-nucleus RNA sequencing on various frozen tissue types, generating a cross-tissue atlas of cellular profiles. By integrating this atlas, we identified shared and tissue-specific features of cell populations, as well as cell types and gene modules associated with neuromuscular, metabolic, and immune components of monogenic diseases and complex traits analyzed by genome-wide association studies.

Aryl hydrocarbon receptor signals in epithelial cells govern the recruitment and location of Helios+ Tregs in the gut

Yusuke Yoshimatsu et al.

Summary: CD4(+) Foxp3(+) regulatory T cells (Tregs) play a crucial role in the homeostasis of the colon, but the mechanism controlling their localization is still unclear. This study demonstrates that indigo naturalis (IN), an aryl hydrocarbon receptor (AhR) agonist, increases the numbers of Helios+ Tregs and MHC class II+ epithelial cells (ECs) in the colon. Additionally, IN treatment leads to an increase in CD25(+) T cells near the surface of ECs in ulcerative colitis (UC) patients compared to other therapies.

Intracellular metabolic adaptation of intraepithelial CD4+CD8αα+ T lymphocytes

Yosuke Harada et al.

Summary: This study revealed the metabolic properties of intestinal intraepithelial lymphocytes (IELs) and found that the intraepithelial compartment becomes hypoxic in the presence of microbes. Induced CD4(+)CD8 alpha alpha+TCR beta(+) T cells (DPIELs) increase in this location and have lower oxygen and glucose consumption, as well as unique alterations in mitochondria.

Comprehensive Profiling of an Aging Immune System Reveals Clonal GZMK+ CD8+ T Cells as Conserved Hallmark of Inflammaging

Denis A. Mogilenko et al.

Summary: This study characterized age-associated alterations in immune cells across multiple mouse organs and identified a distinct population of age-associated GZMK-expressing CD8(+) T cells. These Taa cells, highly clonal with specific features, were found to be a potential target to address age-associated dysfunctions of the immune system. In humans, proportions of the circulating GZMK(+) CDE3(+) T cell population that shares signatures with mouse Taa cells also increased during healthy aging.

Stepwise chromatin and transcriptional acquisition of an intraepithelial lymphocyte program

Mariya London et al.

Summary: The study demonstrates that mesenteric lymph node T cells undergo tissue adaptation upon migrating to intestinal lamina propria and epithelium, with changes in transcription and chromatin. Cells occupying different gut layers have specific CD4 programming, and the transition to intraepithelial lymphocytes involves the shutdown of the T-reg cell program before accessibility to epithelium.

Single-cell chromatin accessibility landscape identifies tissue repair program in human regulatory T cells

Michael Delacher et al.

Summary: Research identified features of human tissue Treg cells with tissue repair functions, which show similarities in gene expression profiles at the single-cell level with murine cells, and shared characteristics with T follicular helper-like cells. Inducing differentiation of human Treg cells into Tfh-like cells can partially replicate the regenerative characteristics of tissue Treg cells.

Single-cell analyses of Crohn's disease tissues reveal intestinal intraepithelial T cells heterogeneity and altered subset distributions

Natalia Jaeger et al.

Summary: Crohn's disease is a chronic transmural inflammation in the gut caused by dysregulated interaction between microbiome and gut immune system. Studies have shown that T cells from intestinal epithelium and lamina propria in CD patients exhibit unique subsets and altered phenotypes during inflammation.
