Summary: In this study, a fish-specific TLR14, MaTLR14, was identified and characterized from Monopterus albus. MaTLR14 plays a critical role in innate immune responses against pathogens. It mainly located in intestinal epithelial cells and can bind to various bacteria and cause agglutination. Further studies showed that MaTLR14 can activate immune responses through MyD88 adaptor and related signaling pathways.

TRIM25 inhibits spring viraemia of carp virus replication by positively regulating RIG-I signaling pathway in common carp (Cyprinus carpio L.)

Summary: In this study, transcriptome analysis was performed to investigate the immune response of common carp against SVCV infection. It was found that the RLR signaling pathway was significantly enriched after SVCV infection. Further investigation revealed that CcRIG-I and CcTRIM25 suppressed the replication of SVCV by inducing the phosphorylation of TBK1, IRF3, and p65, as well as the expression of ifn-1, viperin, isg15, and mx. Additionally, CcTRIM25 was shown to regulate CcRIG-I-mediated immune response through interaction with CARD and promoting K63-linked polyubiquitination of CcRIG-I.

Analysis of Porcine RIG-I Like Receptors Revealed the Positive Regulation of RIG-I and MDA5 by LGP2

Summary: This study characterized the porcine RLR members RIG-I, MDA5, and LGP2 and found that porcine RIG-I and MDA5 exhibited similar signaling activity to distinct dsRNA and viruses compared to their human counterparts. Porcine LGP2 was found to positively regulate RIG-I and MDA5 in porcine alveolar macrophages and exert broad antiviral functions. Interestingly, mutant forms of porcine LGP2 with defects in ATPase and/or dsRNA binding showed constitutive activity likely through RIG-I and MDA5. The research provided significant insights into porcine innate immunity and immune biology.

Interactions Between Commensal Microbiota and Mucosal Immunity in Teleost Fish During Viral Infection With SVCV

TRIF was found to be constitutively expressed in various tissues, with the highest expression in the spleen and lowest in the head kidney. Stimulation with different factors led to upregulation of TRIF, while its downstream signaling involved interactions with TBK1 and activation of interferon-stimulated genes, inflammatory cytokines, and caspase-8-mediated apoptosis.
