Recurrence of Anti-Semaphorin 3B?Mediated Membranous Nephropathy after Kidney Transplantation

Summary: This study reports the first case of early recurrence after transplantation of membranous nephropathy associated with antibodies directed at semaphorin 3B. The findings provide strong evidence that anti-semaphorin 3B antibodies are pathogenic and should be carefully monitored in patients with membranous nephropathy. Furthermore, the study supports the efficacy of rituximab in treating the disease.

Hematopoietic Stem Cell Transplant-M embranous Nephropathy Is Associated with Protocadherin FAT1

Summary: In this study, a novel antigen associated with membranous nephropathy (MN) in patients receiving hematopoietic stem cell transplant (HSCT) was identified using laser microdissection and tandem mass spectrometry (MS/MS). The antigen, called FAT1, was detected in the glomeruli of patients with PLA2R-negative MN. Further studies confirmed the presence of FAT1-associated MN in a majority of HSCT-related MN cases. This study provides valuable insights into the pathogenesis of MN in HSCT patients and may contribute to the development of targeted therapies.

Discovery of anti-Formin-like 1 protein (FMNL1) antibodies in membranous nephropathy and other glomerular diseases

Summary: Evidence has shown that autoantibodies targeting podocytes can cause membranous nephropathy. In this study, a high-density peptide array was used to identify a panel of proteins reactive to sera of membranous nephropathy patients. The protein FMNL1, expressed by macrophages in the tissues of membranous nephropathy patients, was found to have high levels of anti-FMNL1 IgG4 in the sera of these patients. This autoantibody was associated with lack of remission of proteinuria and was also observed in other glomerular disease conditions. These findings suggest a broader role of this autoantibody in different glomerular diseases.

Neural cell adhesion molecule 1 is a novel autoantigen in membranous lupus nephritis

Summary: Research has identified neural cell adhesion molecule 1 (NCAM1) as a target antigen in some cases of membranous lupus nephritis and rare cases of primary membranous nephropathy, colocalizing with IgG in glomerular immune deposits. Circulating antibodies to NCAM1 have been detected in serum of patients with NCAM1-associated membranous nephropathy, suggesting diagnostic significance. Future studies are needed to determine whether anti-NCAM1 antibody levels correlate with disease activity or response to therapy.

Summary: This study found that IMN patients with nephrotic-range proteinuria have significantly higher levels of sPLA2-IB and fewer autophagosomes than those with non-nephrotic-range proteinuria. In vitro experiments demonstrated that sPLA2-IB induced insufficient autophagy in podocytes and promoted podocyte injury through activation of the mTOR/ULK1 signaling pathway. Additionally, inhibition of p38 MAPK abrogated sPLA2-IB-induced autophagy and mTOR/ULK1 activation.
