MYB orchestrates T cell exhaustion and response to checkpoint inhibition

Carlson Tsui et al.

Summary: This study reveals that CD62L(+) T-PEX cells, maintained by the transcription factor MYB, play a crucial role in maintaining the proliferative potential, multipotency, and repopulation capacity of exhausted T cells during chronic infection, thus contributing to long-term antiviral immunity and responsiveness to immunotherapy.

Single-cell antigen-specific landscape of CART infusion product identifies determinants of CD19-positive relapse in patients with ALL

Zhiliang Bai et al.

Summary: The differences in antigen-specific activation states are associated with CD19-positive relapse in ALL patients after CAR-T cell therapy. The deficiency of T helper 2 function and low frequency of early memory T cells are characteristics of relapse. These findings may help predict therapeutic outcomes and guide strategies to improve T cell function for long-term remission.

Side-effect management of chimeric antigen receptor (CAR) T-cell therapy

M-L Schubert et al.

Summary: CAR T-cell therapy targeting CD19-expressing B cells has shown promising results in treating refractory and/or relapsed B-cell malignancies, but also presents challenges such as cytokine release syndrome and neurotoxicity syndrome. Optimal management strategies for the side-effects associated with CAR T-cell therapy are crucial as it expands into other diseases in hematology and oncology.

Single-cell transcriptome analysis of CAR T-cell products reveals subpopulations, stimulation, and exhaustion signatures

Xiaonan Wang et al.

Summary: This study used single-cell transcriptomic analysis to characterize the molecular features of CAR T cell activation, revealing several subpopulations of cells in CAR products with reproducible cellular composition across donors. Targeted data interrogation also showed that a small proportion of antigen-responding CAR-expressing cells exhibited exhaustion signatures with known markers and previously unassociated genes.

Deciphering the Transcriptomic Heterogeneity of Duodenal Coeliac Disease Biopsies

Johannes Wolf et al.

Summary: This study provides a high-resolution analysis of the transcriptomes extracted from duodenal biopsies of children and adolescents with active CD, revealing three distinct CD patient groups. The persistence of symptoms was weakly associated with subgroups, with the CD-high group showing the highest marsh stages and cell cycle rates, indicating virtually complete villous atrophy. Inflammation-level differences between subgroups were further deciphered into immune cell types using cell type de-convolution. The application of self-organizing maps provided high-resolution landscapes of the CD-transcriptome.
