Idebenone Regulates Aβ and LPS-Induced Neurogliosis and Cognitive Function Through Inhibition of NLRP3 Inflammasome/IL-1β Axis Activation

Hyun-ju Lee et al.

Summary: This study explores the effects of idebenone on neuroinflammatory responses, cognitive function, and underlying molecular mechanisms. The results suggest that idebenone can enhance neuroprotective NRF2 expression, reduce neuroinflammation, and improve learning and memory in mice. It also targets NLRP3 inflammasome activation and decreases gliosis and proinflammatory cytokine levels. In a mouse model of Alzheimer's disease, idebenone improves cognitive dysfunction and inhibits neuroinflammation.

NLRP3 inflammasome of microglia promotes A1 astrocyte transformation, neo-neuron decline and cognition impairment in endotoxemia

Ting Xiao et al.

Summary: Infection caused by gram-negative bacteria is a critical global health issue. Advances in techniques such as antibiotics and life-supporting modalities have reduced the mortality rate of infected patients in recent decades. However, infection-related complications, particularly cognitive dysfunction, still have a significant impact on patient mortality and quality of life. This study reveals a novel understanding of the pathogenesis of cognitive dysfunction induced by lipopolysaccharides (LPS) and suggests that the NLRP3 inflammasome in microglia could be a promising target for the treatment of this syndrome.

Neuregulin-1 regulates the conversion of M1/M2 microglia phenotype via ErbB4-dependent inhibition of the NF-κB pathway

Yuqi Ma et al.

Summary: NRG1 inhibits the release of inflammatory factors in microglia and regulates the switching of the M1/M2 microglia phenotype, most likely via ErbB4-dependent inhibition of the NF-kappa B pathway. Exogenous NRG1 treatment or overexpression improved microglial activity and reduced the secretion of the inflammatory factors TNF-alpha and iNOS in vitro. Overall, NRG1 shows promise as a potential therapeutic target for Alzheimer’s disease through its anti-inflammatory properties.

Nilotinib modulates LPS-induced cognitive impairment and neuroinflammatory responses by regulating P38/STAT3 signaling

Jieun Kim et al.

Summary: Nilotinib can modulate neuroinflammatory responses and cognitive function in chronic myelogenous leukemia, by suppressing specific signaling pathways to reduce inflammation levels and improve spatial working memory and cortical dendritic spine number. These findings are important for understanding the impact of leukemia treatment drugs on the nervous system.

Summary: This study found that fatty acid glucoside (FAG) has anti-inflammatory activity and can exert its effects through inhibition of the EGFR/Akt/PI3K pathway. The study demonstrates that FAG is a natural EGFR inhibitor and can simultaneously release NO and inhibit COX, indicating its potential as an anti-inflammatory agent.

miR-155 and miR-146a collectively regulate meningitic Escherichia coli infection-mediated neuroinflammatory responses

Bo Yang et al.

Summary: E. coli infection induced upregulation of miR-155 and miR-146a in astrocytes, which modulated bacteria-triggered neuroinflammatory responses through negative feedback regulation involving the TLR-mediated NF-kappa B and EGFR-NF-kappa B signaling pathways, thus protecting the central nervous system from further neuroinflammatory damage.

The Amyloid-β Pathway in Alzheimer's Disease

Harald Hampel et al.

Summary: Breakthroughs in molecular medicine have highlighted the significance of the amyloid-beta pathway in the pathophysiology of Alzheimer's disease. Established biochemical alterations of the A beta cycle serve as promising targets for the development of disease-modifying therapies. Research indicates a crucial role of A beta pathway dyshomeostasis in the dynamics of AD pathophysiology, supporting the development of A beta-targeting therapeutic strategies for early treatment of AD.

Morphine promotes microglial activation by upregulating the EGFR/ERK signaling pathway

Yaqiong Yang et al.

Summary: This study investigated the regulatory mechanism of epidermal growth factor receptor (EGFR) on microglial activation induced by morphine in BV-2 cells. The results showed that morphine treatment increased CD11b expression, EGFR phosphorylation, ERK1/2 activation, cell migration, and production of proinflammatory cytokines, which were inhibited by blocking the EGFR signaling pathway with AG1478. These findings suggest that the EGFR/ERK signaling pathway could be a novel pharmacological strategy to attenuate morphine tolerance by suppressing microglial activation.

Summary: HER2-positive breast cancer accounts for a significant proportion of cases, with several targeted therapies available including TKIs which have shown promising responses in both early and advanced settings. In patients with central nervous system involvement, TKIs have shown efficacy, which is important for those with limited treatment options and poor prognosis in this setting.
