Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

A Newly Established Cuproptosis-Associated Long Non-Coding RNA Signature for Predicting Prognosis and Indicating Immune Microenvironment Features in Soft Tissue Sarcoma

Jun Han et al.

Summary: This study aimed to determine the prognostic significance of cuproptosis-associated lncRNAs in soft tissue sarcomas (STSs) and their relationship to the tumor immune microenvironment. The study identified a signature of 5 cuproptosis-associated lncRNAs, classified STS patients into high-risk and low-risk groups, and found that high-risk patients had a worse prognosis. A nomogram combining clinical characteristics and risk scores was constructed and showed reliable predictive power. Functional enrichment and tumor immune microenvironment analysis suggested that high-risk STSs tend to be immunologically sensitive tumors.

Connecting copper and cancer: from transition metal signalling to metalloplasia

Eva J. Ge et al.

Summary: Copper is a essential nutrient with both beneficial and toxic properties to cells. Recent progress in transition metal signaling has led to new connections between researchers from different disciplines, with potential for translating basic research into clinical therapies for diseases. The relationship between copper and cancer is particularly important, with opportunities for leveraging disease vulnerabilities through copper-dependent signaling pathways.

Copper metabolism as a unique vulnerability in cancer

Vinit C. Shanbhag et al.

Summary: Over the past 25 years, significant progress has been made in identifying and characterizing proteins that regulate the uptake, intracellular trafficking and export of copper. While copper is essential for growth and development in humans and other mammals, it is also a limiting nutrient for cancer cell growth and proliferation. Oral copper chelators have been shown to have anti-neoplastic and anti-metastatic effects in animals and humans.

Beyond conventional immune-checkpoint inhibition - novel immunotherapies for renal cell carcinoma

David A. Braun et al.

Summary: The management of advanced-stage renal cell carcinoma has been transformed by the development of immune-checkpoint inhibitors (ICIs). However, most patients do not derive durable clinical benefit from these agents. Innovations in immunotherapy for RCC, including ICIs with novel targets, are now in clinical development.

YAP1 Expression in SCLC Defines a Distinct Subtype With T-cell-Inflamed Phenotype

Taofeek K. Owonikoko et al.

Summary: This study identified four subtypes of SCLC using RNA sequencing and immunohistochemistry, with the SCLC-Y subtype associated with an inflamed phenotype and favorable prognosis. Upregulation of interferon-gamma response genes was observed in long-term survivors, with higher expression in patients of the SCLC-Y subtype.

TNF Patterns and Tumor Microenvironment Characterization in Head and Neck Squamous Cell Carcinoma

Qian Long et al.

Summary: By clustering the expression of 46 members of the TNF family, two distinct subtypes with clinical and immune characteristics were identified in HNSCC. A risk scoring system based on the TNF family gene expression profile was constructed, serving as a reliable predictor of overall survival, clinical characteristics, and immune cell infiltration, with potential application as a valuable biomarker for HNSCC immunotherapy.

Primary vulvar squamous cell carcinomas with high T cell infiltration and active immune signaling are potential candidates for neoadjuvant PD-1/PD-L1 immunotherapy

Kim E. Kortekaas et al.

Summary: An active immune signaling profile is present in 35% of primary FIGO I-III VSCCs, suggesting potential responsiveness to neoadjuvant PD-1/PD-L1 immunotherapy. High intraepithelial infiltration with CD4 or CD8 T cells was associated with longer overall and recurrence-free survival, outperforming molecular subtype and disease stage. Strong T cell infiltrated VSCC displayed a coordinated immune response with different lymphocyte and myeloid cell subsets.

Siglec15 shapes a non-inflamed tumor microenvironment and predicts the molecular subtype in bladder cancer

Jiao Hu et al.

Summary: Siglec15 is specifically overexpressed in the tumor microenvironment (TME) of various cancers, likely designing a non-inflamed TME in bladder cancer (BLCA). High expression of Siglec15 in BLCA correlates with lower immune infiltration, lower response to cancer immunotherapy, and a higher incidence of hyperprogression. A combination of anti-Siglec15 and cancer immunotherapy may be a more effective strategy than monotherapy, while an immune risk score (IRS) could accurately predict prognosis and response to treatment.
