Development of bone cell microarrays in microfluidic chips for studying osteocyte-osteoblast communication under fluid flow mechanical loading

Charlotte Yvanoff et al.

Summary: Bone tissue constantly remodels in response to mechanical loads, and disruptions in bone cell activities can lead to disorders like osteoporosis. To understand and develop therapeutic strategies for bone disorders, a microfluidic platform was developed to study the bone remodelling cycle at a molecular level. The platform utilized a cell printing method to create living bone cell arrays in a microfluidic channel, enabling the study of cell-cell communication between osteocytes and osteoblasts. High-resolution characterization of physical contacts was achieved using correlative atomic force microscopy (AFM) and fluorescence microscopy. The platform also demonstrated compatibility with single-cell mechanostimulation and analysis of mechanoresponse.

Mechanically stimulated osteocytes promote the proliferation and migration of breast cancer cells via a potential CXCL1/2 mechanism

Summary: Bone is a common site for breast cancer metastasis, with mechanically stimulated osteocytes releasing factors that enhance cancer cell proliferation and migration. The secretome of mechanically activated osteocytes contains specific cytokines that potentially drive breast cancer metastasis to bone.

Moderate tibial loading and treadmill running, but not overloading, protect adult murine bone from destruction by metastasized breast cancer

Shubo Wang et al.

Summary: Moderate mechanical loading can suppress tumor-induced bone destruction in breast cancer, but higher magnitude loading can accelerate bone destruction. Different exercise regimens have varying effects on bone cells and bone marrow. Optimization of exercise parameters is needed to harness the skeletal benefits of exercise in metastatic breast cancer.

Summary: In this study, a 3D co-culture model was established with primary human osteoblasts, osteoclasts, and osteocytes, providing preliminary information on the performance of bone graft materials by analyzing the interactions of different cell types. The experiment showed typical morphology and gene expression of various markers in the three cell species, as well as enzyme activities relevant to osteoblasts and osteoclasts.

Disrupted osteocyte connectivity and pericellular fluid flow in bone with aging and defective TGF-β signaling

Summary: Skeletal fragility in the elderly is not solely due to bone mass loss, with osteocytes playing a key role in bone aging. The degeneration of the lacunocanalicular network may lead to compromised mechanostimulation of osteocytes, highlighting them as potential targets for age-related therapeutic interventions to improve bone health.

Emerging Players in Prostate Cancer-Bone Niche Communication

Giulia Furesi et al.

Summary: Patients with advanced prostate cancer often develop skeletal metastases that can lead to fractures, disability, and increased mortality. Interactions between tumor cells and osteoblasts are believed to play a major role in the osteotropism of prostate cancer. Prostate cancer-derived extracellular vesicles (EVs) are emerging as key players in reprogramming osteoblasts and supporting the formation of premetastatic niches, explaining the complexity of prostate cancer osteotropism.

Novel in vitro microfluidic platfor for osteocyte mechanotransduction studies

Summary: Osteocytes are the major mechanosensing cells in bone remodeling, and this project aims to design and fabricate a multi-shear stress, co-culture platform to study the interaction between osteocytes and other bone cells under varying flow conditions. By utilizing microfluidic devices, significant differences in RANKL levels between channels with different shear stress levels were observed, showing that pre-osteoclast differentiation is directly affected by adjacent flow-stimulated osteocytes. The platform provides an optimal tool for bone cell mechanistic studies and potential drug target discovery for clinical treatments of bone-related diseases.
