Advanced photodynamic therapy with an engineered M13 phage targeting EGFR: Mitochondrial localization and autophagy induction in ovarian cancer cell lines

Barbara Bortot et al.

Summary: This study proposes a bacteriophage-based receptor targeted photodynamic therapy for treating EGFR-positive ovarian cancer. The modified M13 bacteriophage with an EGFR binding peptide displayed on its surface effectively kills cancer cells through ROS generation by photosensitizers. It also downregulates EGFR expression and induces autophagy.

Functionalized Liposome and Albumin-Based Systems as Carriers for Poorly Water-Soluble Anticancer Drugs: An Updated Review

Sofia Teixeira et al.

Summary: Cancer is a major cause of death worldwide. To overcome the problems associated with chemotherapy, the scientific community has developed functionalized nanosystems to deliver drugs to cancer cells, targeting overexpressed receptors, proteins, and organelles. This review focuses on the recent developments of liposomes and albumin-based nanosystems as carriers for poorly water-soluble drugs.

Fullerenes against COVID-19: Repurposing C60 and C70 to Clog the Active Site of SARS-CoV-2 Protease

Tainah Dorina Marforio et al.

Summary: The persistency of COVID-19 and its variants requires new treatments in addition to vaccines. Computational chemistry suggests that fullerenes and carbon nanomaterials have potential as antiviral agents. This study shows that C-60 has strong binding energy to the main protease of the SARS-CoV-2 virus, M-pro, and may be more effective than the known inhibitor masitinib. The use of C-70 as an innovative scaffold for the inhibition of SARS-CoV-2 M-pro is also proposed.

Improvement of Photodynamic Activity by a Stable System Consisting of a C60 Derivative and Photoantenna in Liposomes

Risako Shimada et al.

Summary: This study investigates the photodynamic activity of a dyad system composed of a lipid membrane-incorporated fullerene derivative and a photoantenna molecule in liposomes. It is found that the release of the fullerene derivative from the liposomes can disrupt the dyad system and reduce its photodynamic activity. The hydrophobicity/hydrophilicity balance of the substituent in the fullerene derivatives is shown to be crucial for obtaining a dyad system with high photodynamic activity.

Reversible Dimerization of Human Serum Albumin

Alexey Chubarov et al.

Summary: The Pulsed Dipolar Spectroscopy (PDS) methods of Electron Paramagnetic Resonance (EPR) were used to study reversible non-covalent dimers of Human Serum Albumin (HSA), showing that chemical modifications of Cys-34 can affect dimerization. The weak non-covalent dimers formed by HSA at physiological concentrations have a well-defined structure and can easily revert back to monomers, playing an important role in transport and binding processes.

Dissecting the Supramolecular Dispersion of Fullerenes by Proteins/Peptides: Amino Acid Ranking and Driving Forces for Binding to C60

Tainah Dorina Marforio et al.

Summary: The study quantitatively investigated the interactions between twenty proteinogenic amino acids and C-60 using molecular dynamics simulations. It was found that the conserved amino acid backbone and the contribution from amino acid side chains played key roles in the binding, with influences from van der Waals, hydrophobic, and polar solvation interactions. The presence of C-60 led to specific configurations and geometric structures in the interacting residues and amino acid backbone.

Summary: Chemotherapeutics are commonly used for cancer treatment, but often have off-target effects on healthy cells. Gene therapy is becoming a trend in oncotherapy, aiming to safely deliver genetic material to target cells using non-viral carriers. Using terpolymer-based nanocapsules decorated with folic acid for active targeting shows potential for more effective cancer therapies.

Human Serum Albumin-Oligothiophene Bioconjugate: A Phototheranostic Platform for Localized Killing of Cancer Cells by Precise Light Activation

Andrea Cantelli et al.

Summary: The development of HSA-oligothiophene bioconjugates allows insoluble oligothiophenes to be used in physiological environments, exhibiting water solubility, biocompatibility, and the ability to produce ROS effectively to eradicate cancer cells. The bioconjugate serves as a theranostic platform for ROS generation and imaging, while also acting as a real-time self-reporting system to monitor apoptosis. Its phototoxicity is localized and precise, demonstrating efficient killing of cancer cells through light activation.
