Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Image-guided thermosensitive liposomes for focused ultrasound enhanced co-delivery of carboplatin and SN-38 against triple negative breast cancer in mice

Summary: Triggerable nanocarriers have the potential to improve the therapeutic index of existing anticancer agents by enabling localized delivery and release of therapeutic cargos. In this study, the first co-delivery of SN-38 and carboplatin was achieved using an MRI-visible thermosensitive liposome, showing both thermosensitivity and MR-imageability. The co-delivery resulted in significant improvements in anti-tumor activity in triple negative breast cancer xenografts tumors in mice.

Most liposomes achieve sufficient circulation half-life through polyethylene glycol surface modification, while DPPG(2) has been identified as a promising alternative. In vitro and in vivo studies suggest that higher mol% DPPG(2) increases Apolipoprotein E adsorption, decreases complement activation, and does not exhibit accelerated blood clearance, potentially offering advantages over PEG-TSL. Additionally, low complement activation for 20-30 mol% DPPG(2)-TSL presents a unique attribute for clinical evaluation.

Ultrasound-triggered herceptin liposomes for breast cancer therapy

Summary: Functionalizing liposomes with monoclonal antibodies, particularly anti-HER2 antibodies, can enhance targeted drug delivery to HER2 positive breast cancer cells. Low frequency ultrasound (LFUS) triggered drug release from liposomes can be further enhanced by the conjugation of Trastuzumab, increasing drug uptake and cellular toxicity in HER2+ cell lines. Combining immunoliposomes with LFUS is a promising technique to improve drug delivery efficiency and reduce cytotoxicity of antineoplastic drugs.
