Nanocarriers for Delivery of Oligonucleotides to the CNS

David Male et al.

Summary: This review discusses the ability of nanoparticles with oligonucleotides to enter the brain and modulate gene expression or disease. The size and surface charge of the nanocarrier, as well as the structure of the oligonucleotide and chemical formulation of the nanocarrier, play important roles in determining their efficacy. Successful gene therapy with oligonucleotides in humans has required intrathecal injection, while in animal models, progress has been made with intraventricular or intravenous injection.

Accumulation of Mitochondrial RPPH1 RNA Is Associated with Cellular Senescence

Ji Won Lee et al.

Summary: Mitochondrial long noncoding RNAs exhibit expression fluctuations during cellular senescence, with RPPH1 RNA possibly interacting with RNA-binding protein AUF1 to accumulate in senescent fibroblast mitochondria, promoting cellular senescence. Partial complementarity of RPPH1 RNA with target mRNAs prevents decay in proliferating fibroblasts, demonstrating a role in mRNA stability and cellular senescence regulation.
