New Biomarker in Chagas Disease: Extracellular Vesicles Isolated from Peripheral Blood in Chronic Chagas Disease Patients Modulate the Human Immune Response

Rafael Pedro Madeira et al.

Summary: Our study focused on characterizing and comparing circulating extracellular vesicles (EVs) in chronic Chagas disease (CCD) patients and controls. We found that CCD patients had lower concentration of circulating EVs, which were able to induce differential production of IFN-gamma and IL-17 in macrophages, especially in earlier stages of the disease, indicating their potential as biomarkers for disease progression monitoring.

Trypanosoma cruzi trans-Sialidase as a Potential Vaccine Target Against Chagas Disease

Kelli Monteiro da Costa et al.

Summary: Chagas' disease, caused by Trypanosoma cruzi, was described by Dr. Carlos Chagas in the early 20th century. One important discovery was trans-sialidase, an enzyme that masks the parasite's presence and dampens the immune response. Research into the disease has identified key events in the biochemical mechanism of T. cruzi-host cell interactions.

Stress Induces Release of Extracellular Vesicles by Trypanosoma cruzi Trypomastigotes

Camilla Ioshida Vasconcelos et al.

Summary: This study evaluated the release and internalization of EVs from Trypanosoma cruzi under different stress conditions. It was found that EV release is dependent on membrane structure and parasite integrity, and stress conditions do not affect the functional properties of EVs during interaction with host cells. EVs released under stress conditions maintained their proinflammatory activity and stimulated the expression of certain genes in preactivated macrophages. Variations in EV release under stress conditions may be a physiological response against environmental changes.

Biophysical and Biochemical Comparison of Extracellular Vesicles Produced by Infective and Non-Infective Stages of Trypanosoma cruzi

Lissette Retana Moreira et al.

Summary: This study characterized the proteomes and nanomechanical properties of extracellular vesicles released by different life stages of Trypanosoma cruzi, the etiologic agent of Chagas disease. The findings revealed significant differences in surface molecular cargos of vesicles between trypomastigotes and epimastigotes, with trypomastigotes exhibiting higher adhesion properties. This suggests a remarkable surface remodeling throughout the parasite's life cycle, potentially impacting the ability of extracellular vesicles to participate in cell communication in diverse infection niches.

Chagas heart disease: An overview of diagnosis, manifestations, treatment, and care

Roberto M. Saraiva et al.

Summary: Chagas heart disease, caused by Trypanosoma cruzi, affects around 30% of patients. It is classified into four stages based on criteria including ECG, echocardiogram, and clinical findings. CHD presents with various clinical manifestations, with sudden cardiac death, heart failure, and stroke being the main complications. Sudden cardiac death is the most common cause of death in CHD patients, followed by heart failure and stroke.

Extracellular vesicles and leishmaniasis: Current knowledge and promising avenues for future development

George Dong et al.

Summary: Extracellular vesicles (EVs) are small membrane-bound delivery vehicles that can induce changes in neighboring cells' physiological status through the transfer of key molecules, potentially playing roles in various pathological processes. Leishmaniasis, caused by Leishmania parasite, serves as an important case study where EVs are studied extensively for their involvement in disease exacerbation and drug resistance. Additionally, EVs are investigated for vaccine development and as potential biomarkers for disease and drug resistance.

Characterization of murine extracellular vesicles and Toxoplasma gondii infection

Marta Marques Maia et al.

Summary: This study characterized extracellular vesicles (EVs) derived from mice infected with Toxoplasma gondii, showing that EVs can play a role in the host-parasite interaction. The study also revealed that miRNAs in EVs are involved in modulating the immune response against T. gondii. These findings suggest that the concentration of EVs could potentially differentiate between infected and non-infected hosts.

The Case for the Development of a Chagas Disease Vaccine: Why? How? When?

Eric Dumonteil et al.

Summary: Chagas disease, transmitted by triatomine insect vectors, has become a global disease with limited drug treatment options. The development of a vaccine is crucial for better disease control. Recent pre-clinical studies have shown progress and challenges in vaccine development, including expanding vaccine platforms, understanding immune protection, and assessing vaccine effects on cardiac function.
