Second Heart Field-Derived Cells Contribute to Angiotensin II-Mediated Ascending Aortopathies

Hisashi Sawada et al.

Summary: The study revealed that SHF-derived cells play a critical role in thoracic aortopathies by maintaining vascular integrity through LRP1 and transforming growth factor-beta signaling associated with increased levels of aortic PAI1.

Embryologic Origin Influences Smooth Muscle Cell Phenotypic Modulation Signatures in Murine Marfan Syndrome Aortic Aneurysm

Albert J. Pedroza et al.

Summary: This study provides a detailed characterization of aortic root smooth muscle cells (SMCs) in Marfan syndrome (MFS), revealing the involvement of SMCs derived from both the second heart field (SHF) and neural crest in the formation of aortic root aneurysms. The study also highlights the subtly distinct transcriptional responses between these two lineages and suggests that TWIST1 may play a key role in the enriched collagen synthesis in SHF-derived SMCs in MFS.

Distinct Contribution of Global and Regional Angiotensin II Type 1a Receptor Inactivation to Amelioration of Aortopathy in Tgfbr1M318R/+ Mice

Emily E. Bramel et al.

Summary: This study investigates the effect of AT1R signaling attenuation on aortic disease in a mouse model of LDS. The results show that both systemic and regional AT1R signaling attenuation can reduce the diameter of the aorta and improve tissue morphology in LDS mice, with the more complete effect observed in germline Agtr1a deletion. However, regional AT1a inactivation in SHF-derived lineages has a more modest effect on aortic diameters and is associated with lower blood pressure.

Vascular Smooth Muscle Cell Subpopulations and Neointimal Formation in Mouse Models of Elastin Insufficiency

Chien-Jung Lin et al.

Summary: This study investigates the mechanism of neointima formation due to Eln insufficiency in a mouse model, showing that cells from the secondary heart field are significant contributors to intimal hyperplasia. Two unique SMC populations were identified in the neointima of SMC-Eln knockout mice, suggesting shared mechanisms with adult vessel injury models.

Summary: This study reveals that a pathogenic TGFBR1 variant causes lineage-specific SMC defects informing the etiology of LDS-associated aortic root aneurysm. As a potential pharmacological strategy, our results highlight a combination treatment with activin A and rapamycin that can rescue the SMC defects caused by the variant.

Excess Provisional Extracellular Matrix: A Common Factor in Bicuspid Aortic Valve Formation

Christine B. Kern

Summary: Bicuspid aortic valve (BAV) is a common cardiovascular malformation without current therapeutic options. Studies on murine BAV models suggest that excess versican within the provisional extracellular matrix (P-ECM) is a common factor in BAV development, potentially serving as therapeutic targets and biomarkers of disease progression.
