Non-canonical function of DGCR8 in DNA double-strand break repair signaling and tumor radioresistance

Summary: In response to DNA double-strand breaks, repair proteins are recruited to the damaged sites through ubiquitin signaling. DGCR8, a microRNA biogenesis factor, promotes tumor resistance to X-ray radiation independently of its Drosha-binding ability. Upon radiation, ATM kinase and USP51 deubiquitinase mediate the activation and stabilization of DGCR8. The interaction of these proteins leads to the recruitment of DGCR8 and RNF168 to sites of damage, promoting histone H2A ubiquitination, repair of DSBs, and radioresistance.
