Role of Wnt signaling and sclerostin in bone and as therapeutic targets in skeletal disorders

Francesca Marini et al.

Summary: Wnt signaling and sclerostin play crucial roles in bone metabolism and skeletal disorders. The use of anti-sclerostin antibodies has been proven to be safe and effective in treating low bone mass phenotypes.

Exosomal circ_0001190 Regulates the Progression of Gastric Cancer via miR-586/SOSTDC1 Axis

Chao Liu et al.

Summary: This study investigated the role of circ_0001190 in gastric cancer (GC) and its regulatory mechanism. The results showed that circ_0001190 levels were downregulated in GC, while miR-586 levels were upregulated. Further functional experiments demonstrated that overexpression of circ_0001190 inhibited cell viability, proliferation, angiogenesis, migration, and invasion, while promoting apoptosis in GC cells. Circ_0001190 acted as a sponge for miR-586 to regulate the expression of SOSTDC1. Additionally, miR-586 promoted the progression of GC by interfering with SOSTDC1. Exosomal circ_0001190 overexpression inhibited GC development through the miR-586/SOSTDC1 axis, suggesting a potential targeted therapy for GC.

Revitalizing mouse diphyodontic dentition formation by inhibiting the sonic hedgehog signaling pathway

Chuanqing Mao et al.

Summary: The study showed that the Shh signaling pathway inhibitor effectively inhibited the expression of Shh signaling pathway components and promoted cell proliferation to revitalize the rudimentary successional dental lamina. Additionally, the formation of diphyodontic dentition may be related to up-regulation of FGF signal and the Sostdc1-Wnt negative feedback loop.

MSX1 Drives Tooth Morphogenesis Through Controlling Wnt Signaling Activity

J-M Lee et al.

Summary: Tooth agenesis is a common birth defect that occurs due to morphogenesis failure in early tooth development. This study shows that Msx1 regulates Wnt signaling by suppressing the expression of Dkk2 and Sfrp2, and that Sostdc1/Wise also plays a role in the odontogenic pathway downstream of Msx1. The findings suggest that modulation of Wnt signaling by Dkk2 and Sostdc1 is crucial for proper tooth morphogenesis.

Identification and validation of plasma biomarkers for diagnosis of breast cancer in South Asian women

Thangarajan Rajkumar et al.

Summary: Breast cancer is the most common malignancy in women globally. The development of a reliable plasma biomarker panel could provide a non-invasive and cost-effective method for population-based screening of the disease. This study identified several candidate protein markers and epigenetic markers for breast cancer diagnosis and validated their effectiveness in differentiating breast cancer from benign diseases and healthy individuals. A combination of protein markers demonstrated moderate sensitivity and specificity, while the methylation status of SOSTDC1, DACT2, and WIF1 showed high sensitivity and specificity in distinguishing breast cancer.

USP15 negatively regulates lung cancer progression through the TRAF6-BECN1 signaling axis for autophagy induction

Mi-Jeong Kim et al.

Summary: The study reveals that USP15 is significantly downregulated in lung cancer patients, and its knockout leads to increased migration, invasion, and autophagy induction in lung cancer cells. USP15 interacts with BECN1 and deubiquitinates it, thereby attenuating autophagy induction. Additionally, the expression of USP15 is associated with the upregulation of certain oncogenes and downregulation of tumor suppressor genes in lung cancer patients.
