Summary: The emerging issues of concern are non-alcoholic fatty liver disease (NAFLD) and its advanced stage non-alcoholic steatohepatitis (NASH), which may lead to chronic liver complications. Oxidative stress and metabolic disorders play important roles in the progression of NASH. Currently, NASH treatment is primarily focused on lifestyle changes due to the lack of approved therapeutic options.

Hepatic stellate cells in physiology and pathology

Summary: Hepatic stellate cells (HSCs) play critical roles in the normal liver and in response to injury. They are activated during liver injury and produce extracellular matrix in liver fibrosis. In the absence of injury, HSCs are in a quiescent state and store retinoids or vitamin A-containing metabolites. They also enhance the inflammatory response and express growth factors crucial for liver development and regeneration. Recent studies have identified diverse phenotypic alterations and unique subpopulations of HSCs.

A Randomized, Controlled Trial of the Pan-PPAR Agonist Lanifibranor in NASH

Summary: In a randomized trial, the pan-PPAR agonist Lanifibranor showed significant improvement in histologic features of NASH at a dose of 1200 mg. However, adverse events such as weight gain, anemia, peripheral edema, diarrhea, and nausea were more common with Lanifibranor compared to placebo.

While cardiovascular disease and extrahepatic malignancy are the main causes of death in NAFLD patients, advanced liver fibrosis is a key prognostic marker for liver-related outcomes and overall mortality.

These genetic abnormalities activate MAPK and PI3K/AKT signaling pathways leading to dedifferentiation of DTCs. Research in recent years has focused on strategies to restore NIS expression and RAI uptake.
