Dexamethasone can attenuate the pulmonary inflammatory response via regulation of the lncH19/miR-324-3p cascade

Ye Chen et al.

Summary: This study investigated the roles of lncRNAs in regulating the pulmonary inflammatory response under dexamethasone treatment. Results indicated that lncH19 was closely associated with the inflammatory response and could potentially regulate miR-324-3p to modulate inflammation. Dex treatment attenuated the inflammatory response by regulating the lncH19/miR-324-3p cascade.

Second-generation lung-on-a-chip with an array of stretchable alveoli made with a biological membrane

Pauline Zamprogno et al.

Summary: A new generation lung-on-a-chip utilizing a biological membrane made of collagen and elastin has been developed to mimic an array of tiny alveoli with in vivo-like dimensions. This membrane outperforms traditional PDMS in terms of stability, biodegradability, and tunability, offering the potential to more accurately reproduce air-blood barrier functions.

Specific cytokines in the inflammatory cytokine storm of patients with COVID-19-associated acute respiratory distress syndrome and extrapulmonary multiple-organ dysfunction

Jiajia Wang et al.

Summary: Levels of inflammatory cytokines IL-6, IL-8, IL-10, and TNF-alpha are associated with the development of ARDS and extrapulmonary MOD in COVID-19 patients. These cytokine levels significantly increased in ARDS patients and correlated with coagulation parameters, DIC, kidney injury, and oxygenation levels. Non-survivors had higher levels of IL-6 and IL-10 compared to survivors.

Bioprintable Lung Extracellular Matrix Hydrogel Scaffolds for 3D Culture of Mesenchymal Stromal Cells

Bryan Falcones et al.

Summary: Mesenchymal stromal cell (MSC) therapy for acute respiratory diseases relies on MSCs' secretion of paracrine factors. Preconditioning the MSCs by growing them in a lung-derived extracellular matrix (ECM) can enhance their therapeutic efficacy. Bioink based on decellularized porcine lung ECM hydrogels supports 3D culturing of lung-resident MSCs without additional crosslinking, leading to improved cell-matrix interactions and increased expression of key receptors. This approach shows promise for enhancing MSC preconditioning for therapeutic applications.

The Role of Pulmonary Surfactants in the Treatment of Acute Respiratory Distress Syndrome in COVID-19

Shengguang Wang et al.

Summary: Lung alveolar type-II cells produce pulmonary surfactant, consisting of proteins and lipids which play crucial roles in preventing atelectasis and respiratory function. The SARS-CoV-2 virus damages these cells, leading to decreased production of pulmonary surfactant and contributing to the development of COVID-19. Clinical trials are underway to evaluate the potential therapeutic benefits of pulmonary surfactant in COVID-19 patients.

Oxygen Biosensors and Control in 3D Physiomimetic Experimental Models

Jorge Otero et al.

Summary: Traditional cell culture is transitioning towards physiomimetic approaches to replicate healthy and pathological cell environments. While novel bioengineering techniques have facilitated the design of tissue-specific in vitro models, the critical limitation of lower oxygen diffusion in 3D culture still requires further research and efforts. Recently, advanced 3D devices with integrated biosensors are introducing promising solutions to better control oxygen delivery to cells and simulate disease conditions involving hypoxia.
