Summary: The study demonstrated that quercetin (QCT), a natural flavonoid, inhibited ferroptosis in renal proximal tubular epithelial cells by reducing lipid ROS levels and increasing glutathione (GSH) levels. QCT also ameliorated acute kidney injury induced by ischemia-reperfusion (I/R) or folic acid (FA). Furthermore, RNA-sequence analysis revealed ATF3 as a key factor in QCT's inhibition of ferroptosis. This approach opens the possibility for further crystal structures of GPX4 in complex with potential cancer drug targets.
