MyD88 Deficiency, but Not Gut Microbiota Depletion, Is Sufficient to Modulate the Blood-Brain Barrier Function in the Mediobasal Hypothalamus

Christina N. Heiss et al.

Summary: This study demonstrates that the blood-brain barrier (BBB) function in the mediobasal hypothalamus (MBH) is not affected by gut microbiota, but MyD88 deficiency can modulate BBB function in the MBH by reducing the number of cells in direct contact with circulation.

ACE2, Circumventricular Organs and the Hypothalamus, and COVID-19

Wei-Yi Ong et al.

Summary: SARS-CoV-2 can enter cells by binding to ACE2, and it may enter the brain through circumventricular organs and the hypothalamus. Binding of the virus with ACE2 could lead to a reduction in the ACE2 / Ang (1-7) / Mas receptor signaling pathway, affecting neural activity and the hypothalamic-pituitary-adrenal axis. The virus may also affect the hypothalamus via transneuronal spread along olfactory/limbic pathways. Exploring potential therapeutic pathways to prevent or reduce neurological symptoms of COVID-19 is important.

Summary: TRPM8 plays a crucial role in activating autonomic hypothalamic neuronal circuits under innocuous and nocuous cold stimuli, affecting sleep, behavior, and neuronal activation. The behaviors of TRPM8 KO mice in response to innocuous and nocuous cold stimuli differ significantly from those of WT mice.

Role of astroglial toll-like receptors (TLRs) in central nervous system infections, injury and neurodegenerative diseases

Lun Li et al.

Summary: The central nervous system innate immunity is crucial in infections, neurodegenerative diseases, and brain or spinal cord injuries, with astrocytes and microglia mediating innate immunity. Toll-like receptors (TLRs) are well-studied pattern recognition receptors, with microglial TLR signaling extensively investigated in CNS pathology. More research is needed to understand the role of astroglial TLRs in diseases and injuries, as they can have both harmful and beneficial effects depending on the local environment.

COVID-19 and Toll-Like Receptor 4 (TLR4): SARS-CoV-2 May Bind and Activate TLR4 to Increase ACE2 Expression, Facilitating Entry and Causing Hyperinflammation

Mohamed M. Aboudounya et al.

Summary: Mortality in COVID-19 is caused by respiratory failure, heart failure, and sepsis/multiorgan failure. TLR4, an innate immune receptor, recognizes viral proteins and triggers immune responses. SARS-CoV-2 binds to TLR4, potentially leading to inflammation and multi-organ injury.
