Glucagon-like peptide-1 receptor agonist, liraglutide, reduces heroin self-administration and drug-induced reinstatement of heroin-seeking behaviour in rats

Joaquin E. Douton et al.

Summary: Drug addiction is a chronic brain disease that is difficult to treat. Using the GLP-1 analogue liraglutide (LIR) can reduce heroin addiction-like behavior without causing significant discomfort or major disruptions in food intake and body weight gain.

Gnas Promoter Hypermethylation in the Basolateral Amygdala Regulates Reconsolidation of Morphine Reward Memory in Rats

Peng Liu et al.

Summary: Impairing reconsolidation can prevent relapse of morphine reward memories. DNA methyltransferases play a crucial role in the reconsolidation of opiate-associated memories, specifically in the basolateral amygdala (BLA), and DNA hypermethylation of certain genes is necessary for the reconsolidation of morphine reward memories.

Inhibition of Glycogen Synthase Kinase 3β Activity in the Basolateral Amygdala Disrupts Reconsolidation and Attenuates Heroin Relapse

Yuanyang Xie et al.

Summary: This study demonstrates that GSK-3 beta activity in the basolateral amygdala (BLA) is required for the reconsolidation of heroin drug memory and inhibiting GSK-3 beta activity in BLA disrupts reconsolidation and attenuates heroin relapse. The results also suggest that the effect of GSK-3 beta inhibition is retrieval-dependent and time-specific. Additionally, the long-term effect of GSK-3 beta treatment leads to a detectable decrease in heroin-seeking behavior lasting at least 28 days.

Low-dose polypharmacology targeting dopamine D1 and D3 receptors reduces cue-induced relapse to heroin seeking in rats

Scott T. Ewing et al.

Summary: The study investigated the effects of coadministration of a D1-receptor partial agonist and a D3-receptor antagonist in heroin-seeking rats. Results showed that the coadministration of low doses of both compounds was more effective in reducing cue-induced relapse than either compound alone, with some evidence of drug synergism.

Summary: Manipulation of cannabinoid type 1 receptor (CB1R) signaling in the basolateral amygdala (BLA) influences the strength of cocaine memories and the response to environmental cues, thereby impacting relapse behavior.

Disrupting Reconsolidation by Systemic Inhibition of mTOR Kinase via Rapamycin Reduces Cocaine-Seeking Behavior

Fushen Zhang et al.

Summary: The study reveals that mTOR inhibition within the reconsolidation time-window impairs reconsolidation of cocaine-associated memory, reduces cocaine-seeking behavior, and prevents relapse. These effects are retrieval-dependent and temporal-specific.

The concurrence of DNA methylation and demethylation is associated with transcription regulation

Jiejun Shi et al.

Summary: The mammalian DNA methylome is regulated by methylation and demethylation processes, which have a direct impact on gene expression. Methylation concurrence, measured by the percentage of unmethylated CpGs within partially methylated reads, is strongly correlated with gene expression and can be used to stratify DNA regions based on chromatin accessibility and gene regulation patterns. Methylation concurrence represents a unique methylation metric important for transcription regulation, distinct from conventional metrics such as average methylation and methylation variation.
