Single nuclei RNA sequencing of the rat AP and NTS following GDF15 treatment

Benjamin C. Reiner et al.

Summary: Using snRNAseq approach, this study identified cell type-specific molecular mechanisms associated with GDF15 signaling in the area postrema (AP) and nucleus tractus solitarius (NTS). The findings provide insights into the gene expression alterations and regulatory mechanisms downstream of GDF15, which may serve as potential targets for future pharmacotherapies.

Transcriptional Alterations in Dorsolateral Prefrontal Cortex and Nucleus Accumbens Implicate Neuroinflammation and Synaptic Remodeling in Opioid Use Disorder

Marianne L. Seney et al.

Summary: The study utilized RNA sequencing to analyze brain regions in subjects with OUD, identifying gene transcription related to synaptic remodeling and neuroinflammation. Microglia were implicated as a potential driver for opioid-induced neuroplasticity, with genetic liabilities for risky behavior, ADHD, and depression found in individuals with OUD.

GLP-1 receptor signaling in the laterodorsal tegmental nucleus attenuates cocaine seeking by activating GABAergic circuits that project to the VTA

Nicole S. Hernandez et al.

Summary: Research has shown that administering the GLP-1R agonist exendin-4 (Ex-4) in the LDTg can significantly reduce cocaine-seeking behavior without affecting other behaviors. Furthermore, selectively activating the NTS-to-LDTg circuits can also reduce cocaine seeking through a GLP-1R-dependent mechanism. Additionally, it was found that GLP-1Rs are primarily expressed on GABAergic neurons in the LDTg and the efficacy of Ex-4 in reducing cocaine seeking relies partly on activating LDTg-to-VTA GABAergic projections.

GIP Receptor Agonism Attenuates GLP-1 Receptor Agonist-Induced Nausea and Emesis in Preclinical Models

Tito Borner et al.

Summary: GIPR signaling blocks emesis and attenuates illness behaviors induced by GLP-1R activation while maintaining reduced food intake, weight loss, and improved glucose tolerance. The hindbrain AP/NTS is crucial for processing GLP-1R ligand-induced effects on food intake and weight suppression, as well as emetic stimuli.

A novel dual agonist of glucagon-like peptide-1 receptors and neuropeptide Y2 receptors attenuates fentanyl taking and seeking in male rats

Yafang Zhang et al.

Summary: Illicit fentanyl use has dramatically increased in the United States, leading to a critical need for effective treatments. Research indicates that GLP-1R agonists can mitigate fentanyl-mediated behaviors, and dual agonists of GLP-1Rs and Y2Rs offer a potential new therapeutic approach for opioid use disorders.

The changing opioid crisis: development, challenges and opportunities

Nora D. Volkow et al.

Summary: The current opioid epidemic in the US is one of the most severe public health crises in the country's history, and addressing it is challenging due to its rapidly changing nature and serious outcomes. Success in addressing the crisis will require advances in basic science, development of more acceptable and effective treatments, and implementation of public health approaches, including prevention. The progress made in addressing this crisis should also advance the science and treatment of other substance use disorders.
