mTOR substrate phosphorylation in growth control

Stefania Battaglioni et al.

Summary: The target of rapamycin (TOR), a highly conserved serine/threonine protein kinase, plays a central role in regulating cell growth and metabolism by responding to nutrients, growth factors, and cellular energy. Forming two distinct complexes, TORC1 and TORC2, TOR signaling activates cell growth by stimulating anabolic metabolism and inhibiting catabolic processes. This comprehensive review focuses on mammalian TOR (mTOR) and discusses the phosphorylation of different substrates by mTORC1 and mTORC2, despite sharing a common catalytic subunit. The conclusion is that these two complexes recruit different substrates to phosphorylate a common, minimal motif.

Unlocking mammalian regeneration through hypoxia inducible factor one alpha signaling

Kelsey G. DeFrates et al.

Summary: Traditional approaches in regenerative medicine face limitations in translation and commercialization, prompting the development of therapies that stimulate endogenous processes for regeneration. Recent studies highlight the potential of oxygen-sensing pathways, with upregulation of HIF-1α showing promise in modulating cell metabolism and plasticity for tissue regeneration.

Current Status of Angiogenic Cell Therapy and Related Strategies Applied in Critical Limb Ischemia

Lucia Beltran-Camacho et al.

Summary: Critical limb ischemia is the most severe form of peripheral arterial disease, commonly caused by atherosclerosis leading to progressive blockage of arterial vessels. For patients not suitable for surgical intervention, angiogenic cell therapy has emerged as a promising alternative, showing potential for enhancing revascularization by promoting vessel formation and blood flow recovery.

Small extracellular vesicles containing miR-486-5p promote angiogenesis after myocardial infarction in mice and nonhuman primates

Qingju Li et al.

Summary: This study highlights the key role of sEV miR-486-5p in promoting cardiac angiogenesis. miR-486-5p acts on the fibroblastic MMP19-VEGFA signaling pathway to promote cardiac recovery. Delivery of miR-486-5p-engineered sEVs safely enhanced angiogenesis and cardiac function in a nonhuman primate (NHP) MI model, potentially promoting cardiac repair.
