The Chinese pine genome and methylome unveil key features of conifer evolution

Shihui Niu et al.

Summary: This study provides important resources and insights into conifer evolution and further research on conifer adaptation and development. The researchers found that the large genome size of conifers is mainly due to intergenic regions and transposable element content. Large genes with long introns exhibit higher expression levels. Additionally, the expansion of gene families in conifers is mainly related to stress responses.

Pine species determine fungal microbiome composition in a common garden experiment

Carmen Romeralo et al.

Summary: The composition and abundance of fungal communities in trees are influenced by the host species, as well as the presence of a fungal pathogen. There are distinct core and satellite fungal communities, and the presence of the pathogen does not co-occur with a specific fungal community. Host species play an important role in shaping microbiome assembly in common environments.

New insights into the role of constitutive bacterial rhizobiome and phenolic compounds in two Pinus spp. with contrasting susceptibility to pine pitch canker

Frederico Leitao et al.

Summary: The rhizobiome plays a crucial role in plant health and breeding strategies. This study examines the bacterial rhizobiome of susceptible and resistant pine species and its response to F. circinatum inoculation. The study finds that the rhizobiome of resistant species contains a higher abundance of bacteria associated with disease protection, and these differences are paralleled by a distinct content of phenolic compounds.

Mass spectrometry-based forest tree metabolomics

Ana Margarida Rodrigues et al.

Summary: Research in forest tree species has been slow due to long life cycles, large genome sizes, and lack of genomic tools. Mass spectrometry is used to study metabolite responses to stresses in trees like Pinus, Quercus, and Eucalyptus, but expertise is needed to handle the large and complex datasets in accordance with FAIR principles.

Ras2 is important for growth and pathogenicity in Fusarium circinatum

M. M. Phasha et al.

Summary: This study demonstrated the important role of the Ras2 gene in both growth and pathogenicity of F. circinatum. Knockout mutants showed smaller lesions and colonies, delayed conidial germination, but produced more macroconidia compared to complement mutants and wild type strains. Further research will focus on identifying the pathway(s) through which Ras2 controls these traits.

Dual RNA-Sequencing Analysis of Resistant (Pinus pinea) and Susceptible (Pinus radiata) Hosts during Fusarium circinatum Challenge

Cristina Zamora-Ballesteros et al.

Summary: This study identified differences in molecular responses and physiological traits between the highly susceptible Pinus radiata and the highly resistant Pinus pinea to Fusarium circinatum at an early infection stage. While no significant changes were found in physiological analysis upon pathogen infection, transcriptional reprogramming was observed mainly in the resistant species.

Deciphering the effect of FUB1 disruption on fusaric acid production and pathogenicity in Fusarium circinatum

M. M. Phasha et al.

Summary: This study investigated the potential role of fusaric acid (FA) in the pathogenicity of Fusarium circinatum on pine trees. The disruption of the FUB1 gene was found to affect FA production and contribute to the aggressiveness of the fungus on Pinus patula. These results provide insight into the molecular basis of pathogenicity in this fungus.

Comparative proteomics of Pinus-Fusarium circinatum interactions reveal metabolic clues to biotic stress resistance

Joana Amaral et al.

Summary: This study investigated the response of different host species with varying susceptibility levels to Fusarium circinatum-induced pine pitch canker, revealing core pathways regulating these contrasting responses. The susceptible species may target secondary metabolism and chloroplast redox proteins to favor amino acid production, while the resistant species may rely on chloroplast redox regulation to maintain redox homeostasis. Membrane trafficking-related proteins exclusively found in the resistant species likely explain its defense response against the pathogen.

Temporal physiological response of pine to Fusarium circinatum infection is dependent on host susceptibility level: the role of ABA catabolism

Joana Amaral et al.

Summary: This study explored the response of Pinus radiata and Pinus pinea to Fusarium circinatum infection, revealing different hormonal and oxidative stress-related changes. Pinus pinea showed better resistance to PPC at 6 days post-infection, which could be partly explained by hormone confocal analysis and photosynthesis maintenance for defense mechanism fuel. These results are important for developing hormone-based breeding strategies and inducing resistance to F. circinatum infection.
