Exploration of flashing light interaction effect on improving biomass, protein, and pigments production in photosynthetic bacteria wastewater treatment

Summary: This study investigated how flashing light can enhance bioresource production from the photoheterotrophic organism PNSB. The results showed that certain flashing lights can increase the biomass, protein, carotenoid, and bacteriochlorophyll concentration of PNSB. Luminous light, duty cycle, and frequency were found to be the most important factors affecting biomass, protein, and pigment promotion, respectively. Flashing light also alleviated photoinhibition and increased photosynthetic efficiency, resulting in enhanced biomass accumulation. Furthermore, flashing light reduced energy consumption compared to constant light. Flashing lights may have potential applications in PNSB-based industries for promoting bioresource production.

Summary: In this study, the carotenoid isomerase gene (BnaCRTISO) in rapeseed was edited using the CRISPR/Cas9 system. The mutation of BnaCRTISO significantly affected the petal and leaf color by reducing carotenoid content and increasing the content of some carotenes. Transcriptome analysis indicated that the mutation of BnaCRTISO also affected the expression of important genes involved in carotenoid and flavonoid synthesis pathways.

An improved method for the separation of carotenoids and carotenoid isomers by liquid chromatography-mass spectrometry

Summary: Carotenoids are complex compounds composed of highly conjugated isoprene units that are difficult to identify and quantify due to their extreme hydrophobicity. A new method using a C18 column and isopropanol as eluent successfully separated 23 carotenoid standards and differentiated three groups of isomeric carotenoids. The methodology showed improved compatibility with different categories of carotenoids and the ability to resolve isomeric carotenes, crucial for both assessing carotenoid species and tracing their metabolic pathways.

Deconvoluting apocarotenoid-mediated retrograde signaling networks regulating plastid translation and leaf development

Summary: This study reveals the molecular events causing the leaf phenotype associated with the accumulation of ACS1, which includes two consecutive retrograde signaling cascades. Firstly, ACS1 originating in the plastid drives inhibition of plastid translation (IPT) via nuclear transcriptome remodeling of chlororibosomal proteins, requiring light as an essential component. Subsequently, IPT results in leaf morphological defects via a GUN1-dependent pathway shared with seedlings undergoing chemical IPT treatments and is restricted to an early window of the leaf development.
