Development of a VP2-based real-time fluorescent reverse transcription recombinase-aided amplification assay to rapidly detect Senecavirus A

Summary: In this study, a real-time fluorescent reverse transcription recombinase-aided amplification (rRT-RAA) assay was developed for the detection of Senecavirus A (SVA). The assay showed high sensitivity and specificity, comparable to the previously used PCR assay. Testing results using 189 clinical swine samples demonstrated good consistency and correlation between the rRT-RAA and rRT-PCR assays.

LAMP assay coupled with CRISPR/Cas12a system for portable detection of African swine fever virus

Summary: A method for the convenient, portable, low cost, highly sensitive and specific detection of African swine fever virus (ASFV) was established using a combined LAMP-CRISPR assay. The detection limit reached 7 copies/microliter of p72 gene per reaction with no cross-reactivity with other porcine viruses. The performance of this method showed good consistency with real-time qPCR tests, demonstrating a great potential for on-site monitoring of ASFV in the field.

Onsite detection of plant viruses using isothermal amplification assays

Summary: Plant diseases caused by viruses have a significant impact on crop production and quality, resulting in substantial losses. The options for managing viruses are limited due to their nature as systemic obligate parasites. In this regard, sensitive, robust, and affordable diagnostic assays are needed to detect the presence of viruses in plant materials and prevent their spread. Traditional methods based on biological and physical properties have limitations in terms of sensitivity and time consumption. On the other hand, assays based on viral proteins and nucleic acids offer more specificity, sensitivity, and rapidity. However, they often require sophisticated equipment and technical skills in laboratory settings. Isothermal-based assays, such as LAMP and RPA, provide a simple and reliable alternative that does not require specialized equipment or skills. These assays can be performed using lateral flow devices, making them suitable for onsite detection or testing in the field. Isothermal amplification assays can also be combined with CRISPR/Cas technology to overcome non-specific amplification and cross-contamination issues. The collateral activity associated with some CRISPR/Cas systems has been successfully utilized for the visual detection of plant viruses.

Comparing historical and contemporary SVA isolates in growing pigs revealed differences in cross-neutralizing antibody levels, but similar clinical presentation of vesicular disease.
